OBJECTIVE:To observe the effect of Wulong-pills on the content of complement C3in the blood serum and the expression of mast cell and eosinophil in nasal mucosa of rat models with Spleen deficiency AR,and identify the mechanism of pharmacodynamic action.METHODS:48SPF SD rats were randomly divided into six groups, which one had8rats.The6groups are involved which are the Normal group,the Spleen deficiency AR group(the Control group),the Spleen deficiency AR Wulong-Pills high-does group, the Spleen deficiency AR Wulong-Pills medium-does group, the Spleen deficiency AR Wulong-Pills low-does group, and the Spleen deficiency AR Loratadine group.Establish the rat’s Spleen deficiency model with cold cathartic according to Qiu Baoshan’s report, AR model according to An Yunfang’s. After the model was established, stop putting the rhubarb immersion fluid into stomach,and then give the Wulong-pills or Loratadine to it’s rats by enema for seven days.And then, one hour after the medicine administration,the rats were stimulating by0.5%VOA.Meanwhile, to observe the symptoms especially nasal symptoms. within30minutes after excitation focused on nasal.Drug administration seven days later,the assistant detected the content of complement C3in the blood serum and the expression of mast cell and eosinophil in nasal mucosa.RESULTS:The typical symptoms:Average accumulated points of the typical symptoms was more than5in the model group. The eosinophil in nasal mucosa:The Normal group compared with the Control group that was significant difference (p<0.05).The other groups compared with the Control group that was not significant difference (p>0.05).The mast cell in nasal mucosa:The Normal group compared with the Control group that was significant difference (p<0.05).The Spleen deficiency AR Wulong-Pills high-does group and the Spleen deficiency AR Loratadine group compared with the Control group that was significant difference (p<0.05). The Spleen deficiency AR Wulong-Pills medium-does group and the Spleen deficiency AR Wulong-Pills low-does group compared with the Control group that was not significant difference (p>0.05). The blood complement C3:The Control group has the lower content of the blood complement C3,and compared with the Normal group that was significant difference(p<0.05).The Spleen deficiency AR Wulong-Pills high-does group, the Spleen deficiency AR Loratadine group and the Spleen deficiency AR Loratadine group compared with the Control group that was significant difference (p<0.05),and also compared with the Normal group that was significant difference (p<0.05).The Spleen deficiency AR Wulong-Pills low-does group compared with the Control group that was not significant difference (p>0.05).CONCLUSION:The result indicates that the content of complement C3in the blood serum of the rat models with Spleen deficiency AR is descend.The Wulong-pills can rise the blood Component C3,and reduce the anaphylatoxin generation, and decrease the mast cell and eosinophil in nasal mucosa.Moreover,it maybe restrain the Combination between the blood Component C3and the inflammatory cells in nasal mucosa.So it can reduce the the symptoms especially nasal symptoms of AR,and also the therapeutic effect of Wulong-Pills high-does group is the best.
