Objective:To investigate the carotid artery calcification and ischemic stroke (TIA and cerebral infarction) relationship.Methods:Polymerase chain reaction and technique of restriction fragment length polymorphism were used to detect the genotypes of OPG950and OPG1217in222patients with carotid artery calcification and129patients without carotid artery calcification through CT angiography of head and neck.Results:The serum levels in Carotid artery calcification group is higher than no-Carotid artery calcification group.There is a statistically significant difference between the two groups (t=2.67,P=0.02<0.05), OPG gene T950C polymorphism was existing in in Chinese population of Shandong population, the gene type respectively is TT,TC,CC;and the TT gene type Has the highest percentage. Three genotypes and gene frequency have not Statistical significance between male and female (x2=1.70P=0.43>0.05; x2=0.93P=0.34>0.05); In calcification group CC genotype and gene frequency are higher than no-calcification group, There are Separately Statistical significance (x2=9.11P=0.01<0.05;x2=11.32, P=0.001<0.05); Multivariate Logistic regression was used to adjust for age, sex, smoking, hypertension, C-Reactive protein, Serum calcium,serum phosphate, the calcium-phosphorus product and other risk factors, the incidence of Carotid artery calcification in the person with CC gene genotypes was twice as much as the other gene type。 In calcification group, we find that the serum level with CC gene type is higher than the other group with TT OR TC gene type, There are Separately Statistical significance between the two groups (t=-3.61, P=0.002<0.05),however in no-calcification group,we have not find the difference between the CC genotype group and non-CC genotype group (t=-0.99, P=0.33>0.05).OPG-1217sites also has three genetic type and respectively is TT, TC, CC, with the highest percentage of CC, each genotype and gene frequency are not Significant difference between male and female (x2=0.29P=0.86>0.05; x2=0.10P=0.75>0.05), in the comparison of calcification group and no-calcification group, the genotype and allele frequency were no statistically significant differences(x2=0.68, P=0.71>0.05; x2=0.45, P=0.50>0.05)Conclusions:The serum concentration is higher in calcification group than no-calcification group, it maybe a compensatory mechanism for pretect itself. The ratio of OPG-950CC genotype in calcification group is higher than no-calcification group, C alleles or CC genotypes may be the dangerous genes of carotid artery calcification. And the significantly higher serum concentration with OPG-950CC genotype can be a sign as forecasting risk of calcification. OPG-1217sites may has no effect on calcification.
