Causes major is suddenly or chronic strain of lumbar sprain, sending to young adults, can also be seen in the elderly. In the third lumbar vertebra transverse tip touches the obvious pain points, and can of del Monte and strips and nodular. Is a waist of chronic soft tissue injury diseases, clinical use of conservative treatment of this disease, the effect is not good enough. Acupotomology on the etiology, pathology of chronic soft tissue injury diseases have a new understanding, presents new theories of the etiology of chronic injury of soft tissue pathology. Applying this theory, the use of needle knife solution in the treatment of this disease, has made good on the clinical effect of treatment, but its action mechanism, there is no in-depth study.Objective:By observing the effect of acupotom on morphology changes of local soft tissue, changes in pain threshold, as well as the contents of monoamine neurotransmitters in spinal cord and the hypothalamus of rats with the third lumbar transverse process syndrome, to explore mechanism of acupotom’s effect.Methods:Male SD rats were randomly divided into four groups:normal group, model group, electro-acupuncture (EA) group and acupotom group, the last three groups after were modeled by an operation, EA group and acupotom group were treated respectively.56days after modeled, observed the morphology changes of local soft tissue through HE and detected monoamine neurotransmitter content of hypothalamus and spinal cord with RP-HPLCResults:1. Compared with normal group, model rats’partial muscle was with a large number of inflammatory cell infiltration of inflammatory response in muscle tissue, the muscle fiber cells surrounding inflammatory area, some are faults, some are disordered, and unclear structure, we can see that myosin-concretion, atrophy, edema, degeneration and other changes, the distance between the muscle fibers became greater, edema also existed. In Electro-acupuncture group,the amount of the mononuclear cell infiltration in the center of focus of infection was less than the model group, there was also a muscle tissue inflammatory reaction, but the shrinked muscle cells was less than the model group, and muscle edema also became better. The muscle tissue inflammation around inflammatory area was lightest in the acupotom group, muscle fibers are also more tidy and edema of muscular was not obvious, the number of vascular proliferation between muscle fiber cells was more other three groups.2. The content of5-HIAA of the spinal cord in EA group, acupotom group significantly higher than the model group (P<0.05). But compared with the normal group, there was no significant difference (P>0.05); the content of DOPAC in the hypothalamus in the EA group were more than the other three groups (P<0.05). The content of DOPAC in the spinal cord in acupotom group was significantly lower than the other three groups (P<0.05). The content of5-HT, NE, DA in both the spinal cord and hypothalamus were no significant difference between four groups (P>0.05).Conclusion:1. The model of The third lumbar transverse process syndrome that established by our experiment can simulate the fundamental present symptoms and morphology changes of local soft tissue in the clinic, has some reference value.2. Acupotom’s analgesic effects on the rat model may through the promotion of some monoamine neurotransmitter’s metabolism and liberation. The effect of acupotom on the contents of monoamine neurotransmitter is not the main final way to control the pain, it may probably a component element, which can regulate the contents of other related pain substances through the change of monoamine neurotransmitter to control the pain. |
