Objective To study the expression and role of E-cadherin、 β-catenin and PTEN in cervical squamous cell carcinoma and cervical intraepithelial neoplasia and their relationship with clinicapathologic features.Methods The cervix tissue from75cases of cervical squamous cell carcinoma which were further defined according to the operative findings (FIGO2009) as stage I (27cases), stage Ⅱ (36cases), stage Ⅲ12cases) and graded as high grade (25cases) and moderate grade (34cases) and low grade (16cases),30cases of cervical intraepithelial neoplasia which were subdivided(according to the WHO Scully et al suggestions-1994) as CIN Ⅰ (14cases), CIN Ⅱ-Ⅲ (16cases),and10cases of normal cervix tissue were collected from patients at Laiyang center Hospital of Yantai City from May2005to May2010.The expression of β-Catenin and APC protein was detected using immunohistochemistry.Expression of E-cad、β-catenin and PTEN was examined by using immunohistochemical staining (PV-90002step method) in75cervical squamous carcinoma,30cervical intraepithelial neoplasia (CIN) and10normal cervical epithelia.Results1In normal cervical epithelia tissue, E-cad andβ-catenin showed positive expression at the membrane. In normal cervical epithelia,CIN, and cervical squamous carcinoma specimens,the positive expression rate of E-cad were90%,66.67%、37.33%, and the positive expression rate of E-cad in CIN I,CIN Ⅱ-Ⅲ were85.71%(12/14),50%(8/16); the aberrant high expression rate of β-catenin were10%,46.67%,84%, and the positive expression rate of β-catenin in CIN Ⅰ,CINⅡ-Ⅲ were28.57%(4/14),68.75%(11/16);the positive expression rate of PTEN were90%,70%、40%, and the positive expression rate of PTEN in CIN Ⅰ,CINⅡ~Ⅲ were85.71%(12/14),43.75%(7/16); There were significant differences among the expression of all of them in normal cervical epithelia,CINⅠ and CINⅡ~Ⅲ, cervical squamous carcinoma(p<0.05). There also was significant difference beween the aberrant high expression rate of β-catenin in CIN Ⅱ~Ⅲand cervical squamous carcinoma(P<0.05).2Positive expression rate of E-cad decreased with the increase in grade1to3, and stage Ⅰ toⅡ, it was higher in lymph node metastasis group than that without lymph node matastasis group(P<0.05), It was similar with the positive expression of PTEN.The aberrant expression rate of β-catenin increased with the increase in degree of CIN and development of cervical squamous carcinoma, and the rate in lymph node metastasis group was lower than that without lymph node matasis group, but there were no significant difference in all of them(P>0.05).3Positive correlation between E-cad and PTEN protein expression(P=0.000), negative correlation of the decreased expression of E-cad and the aberrant high expression of β-catenin, and negative correlation of the decreased expression of PTEN and the aberrant high expression of β-catenin were found in cervical squamous carcinoma(p=0.002,P=0.006).Conclusions1. Decreased expression of E-cad, PTEN, and high expression of β-catenin happen in the carcinogenesis.2. aberrant expression of E-cad,PTEN,and β-catenin may have an important role in the carcinogenesis.
