ObjectiveFor further understanding of biological information, judicious evaluation of prognosis for meningioma patient, and further treatment options of the patient, the expression of E-cadherin and β-catenin in meningioma was analyzed with immunohistochemical methods on 49 meningioma specimens in relationship with WHO2000 grading.Material and MethodThe specimens included in this study were collected form 49 meningioma cases in Dept Neurosurg Sir Run Run Shaw Hospital from January 2003 to September 2005. Pathological diagnosis was confirmed with HE staining and two pathologists respectively. 41 cases were classified as grade I (WHO 2000), 3 cases were grade II and 5 cases grade III respectively.All specimens were fixed in formalin, embedded in paraffin and cut into 4μm in continuous sections. En Vision were used in Immunohistochemical staining. Anti- E-cadherin primary antibody was form Gene Tech Shanghai Company Limited (clone 361210, 1:50), Anti-β-catenin primary antibody was form Gene Tech shanghai Company (clone 353910,1:200).The results were graded depending on the positive ratio and intensity of the immunoreactivity. The percentage of immunoreactive tumor cells was scored as: 0(positivecells <5%); 1(positive cells 5%~25%); 2(positive cells 26%~50%); 3(positive cells 51%~75%); 4(>75%). The intensity of the immunoreactive products was scored as: 0(no expression); 1(yellowish); 2(imperial yellow); 3(brown). These two scores were weighted as the immunostaining score. 0: ( —) ; 1-2: ( + ) ; 3-6: (++);>6: (+ + +). Data was analyzed by Chi-square test with SPSS(10.0).ResultsThe negative expression of E-adhesion in meningioma WHO I , II, III was 7.31%, 66.67%, 100.00%, respectively ,(P_e<0.005); The negative expression of β-catenin in meningioma WHO I , II, III was 17.07%, 66.67%, 80.00%, respectively, (P_β<0.05).The negative expression of E-adhesion in meningioma with peritumoral edema 0 , 1, 2, 3 was 6.25%, 14.29%, 40.00%, 100.00%, respectively , (P_E<0.025); The negative expression of p-catenin in meningioma with peritumoral edema 0,1,2,3 was 12.50%, 14.29%, 70.00%, 100.00%, respectively , ( P_β<0.005).The negative expression of E-adhesion in meningioma without invasion (5.88%) was lower than those with invasion (53.33%)(P_e<0.005), the difference was of statistical significant. The difference between negative expression of β-catenin in meningioma without invasion (11.76%) and those with invasion (66.67%)( P_β<0.01) was statically significant, too.The higher negative expression of E-adhesion rate (66.67%) and the negative expression of β-catenin (88.89%) of the meningioma, it was more liable to recur in this series, the difference was of statically significant.Conclusions1. The degree of the expression of E-adhesion and β-catenin was in close correlation with the WHO2000 grading. In the atypical or anaplastic meningioma, the expression of E-adhesion and β-catenin were lower, the difference was of statically significant.2. The degree of the expression of E-adhesion and β-catenin was in close correlation with the peritumoral edema of meningioma. When the less the expression of E-adhesion andβ-catenin, the more possibly peritumoral edema of meningiomas will be.3. The degree of the expression of E-adhesion and β-catenin was in close correlation with the aggressiveness of meningioma. The less expression of E-adhesion and β-catenin, the meningioma will possibly be more invasive.4. The degree of the expression of E-adhesion and β-catenin was in close correlation with the recurrence of meningioma. The less expression of E-adhesion and β-catenin, the meningioma will be more easily to recur.
