The high blood lipids is an important factor to cause cardiovascular and cerebrovascular diseases,which is gaining a high incidence in an ascending trend year by year.The traditional Chinese medicine is rich of functional types and content with little toxic side,an evident effect, and unique advantages in reduing blood lipids.To screen out the traditional Chinese medicine and active parts of reducing blood lipids is of great practical significance to developing Hypolipidemic drugs.The main contents of this paper are:(1) A HPLC method was established for the determination of 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA)reductase inhibitor activity.Lovastatin was considered as positive control,the HPLC method was employer to screen the traditional Chinese medicine:Apocynum venetum,Polygonatum odoratum,Rosa laevigata,Sophora japonica,Rheum palmatum, Crataegus pinnatifida,Pueraria lobata,Typha angustifolia,Alisma orientalis,Polygonatum sibiricum,Chrysanthemum morifolium,Carthamus tinctorius,Ginkgo biloba,Lonicera japonica, Lycium barbarum.(2) To extract and purify the active fraction of the extract of Crataegus pinnatifida with maximum inhibition HMG-CoA reductase activity,and optimize the extraction condition,and compare the inhibition rate of extract and active site of Crataegus pinnatifida to HMG-CoA reductase activity.(3) To identify and assay the ingredient of the active site of Crataegus pinnatifida according to different chemical and spectrum experiment.(4) To make hyperlipemia mode and study on blood lipids regulating function of the active site of Crataegus pinnatifida by animal experiment,Xuezhikang was considered as positive control.The experiment results are as follows: (1) Polygonatum odoratum,Rosa laevigata,Rheum palmatum,Polygonatum sibiricum, Ginkgo biloba,Carthamus tinctorius have no effect on inhibiting HMG-CoA reductase activity, and Apocynum venetum,Sophora japonica,Lycium barbarum,Pueraria lobata,Typha angustifolia,Alisma orientalis,Chrysanthemum morifolium,Lonicera japonica have some effect on inhibiting HMG-CoA reductase activity,and Crataegus pinnatifida can well inhibit HMG-CoA reductase activity,there is no significant deviation to compare with positive control-Lovastatin.(2) The optimum conditions for the extract from Fructus Crataegi:75%ethanol as the solvent,the ratio of material to solvent of 1:8,30 min by supersonic wave,and extraction 2 times. With macroreticular adsorbents of D101 type,the ratio of extracts to resin of 1:6(m/v),the fraction eluted with 60%methanol and 80%methanol is the active site,and it can well inhibit HMG-CoA reductase activity.The 50%inhibiting concentration of extract and active site of Fructus Crataegi respectively is 12.5 mg/mL and 1.1 mg/mL,the inhibition ratio both arising with the increase of concentration.(3) The main ingredient of active site from Fructus Crataegi is the total Flavoniods with the purity of 84.4%.(4) Different dosages of active site of Fructus Crataegi have an evident effect on lowering total cholesterol(TC),triglyceride(TG) and low-density lipoprotein-cholesterol(LDL-C) content, and the effect depends on concentration.At the same time,they have an obvious effect on the elvation of high-density lipoprotein-cholesterol(HDL-C).content,but the effect doesn’t depend on concentration.
