Object To explore the adjust function of Tripterygium wilfordii Hook.f.on MCF-7 cells apoptosis and ER ?/?,preliminary probe into the mechanism of treating SLE.Methods the inhibitory effects of triptolide on MCF-7 cells measured by mtt assay.flow cytometry detection were used to determine triptolide on influence on the activity of MCF-7 cells growth and apoptosis,ER?/? mRNA level were detected by realtime PCR while the protein expression of Caspase-3 level were measured by Western-blot.Results Through the MTT method to detect the lng/mL of triptolide cell inhibitory rate is 6.836%,3ng/mL cell inhibitory rate is 14.519%,lOng/mL cell inhibitory rate is 26.598%,30ng/mL cell inhibition rate is 37.023%.We choose lOng/mL as the drug concentration in this experiment.Triptolide can promote the apoptosis of early and late apoptotic MCF-7 cells,compared with the control group significantly(P<0.05);triptolide intervention,MCF-7 cell ERa mRNA protein expression level decreased,compared with the control group,the difference was significant(P<0.05),while the ER? mRNA protein expression level increased,compared with the control group,significant difference(P<0.05);and the Caspase-3 protein expression increased,compared with the control group,the difference was significant(P<0.05).Conclusion Tripterygium wilfordii can inhibit MCF-7 cell proliferation,and effectively promote the apoptosis;reducing the ERa mRNA,increasing expression of ERP mRNA,and inhibiting the ER transcription activity in the signaling pathway so as to achievethe effect of SLE treatment.
