Objective: The research Ehuang Decoction retention enema in treatment of coloncancer SW480expression in transplanted tumor of human E-cadherin,Toinvestigate the expression of E-cadherin in tumor growth, invasion,metastasis of mechanism, and further provide theoretical basis for thetreatment of malignant tumor of digestive tract.Material and methods: Choose healthy SPF2BALB/c mice,female,age4-6weeks,body weight of20±2g;40healthy SD rats,weighing(180±15)g.InoculatingSW480colon cancer orthotopic transplantation tumor,after the success ofmodeling,according to the random number table grouping method40healthy SDrats were divided into4groups,with10rats in each group:group A:modelcontrol group;group B:positive control group;group C:Ehuang Decoction highdose group;group D:Ehuang Decoction low dose group.Fasting24h afterinoculation,drug treatment to normal feed water7d.Group A:model controlgroup,with sterile water for injection by10ml/kg·QD enema;group B:positivecontrol group,intraperitoneal injection of5-FU,the dosage is50mg/kg·QOD;group C:Ehaung Decoction high dose group,2ml(8g)/qd·only weightenema;group D:Curcuma Decoction low dose group,2ml(0.8g)/qd·only weightenema.Continuous administration of14d in each group were decapitated therats,dissected and the tumor tissue weighing,removal of thymus,spleenweight,inhibition rate,thymus index,spleen index were calculated.Analysis ofE-cadherin expression in transplanted tumor tissues by EnvisionTMtwo stepmethod,to investigate the effect of Ehaung Decoction on human colorectalcancer SW480xenograft tumor invasion,metastasis,and the mechanism isdiscussed.Results:1, Ehuang Decoction can improve the general living conditions of SW480transplanted tumor in rats; 2, Ehuang Decoction can inhibit tumor growth of SW480xenograft tumorrats;3, Ehuang Decoction high and low dose group inhibition rates were26.59%、15.51%;4, Ehuang Decoction can improve the body’s immune function, enhance theability of anti-tumor, can induce the rat thymus, spleen growth;5, Ehuang Decoction can to some extent reduce the missing SW480xenografttumor E-cadherin expression, thus reducing the possibility of livermetastases occur;6, Ehuang Decoction high dose group and positive control group in similarimpact SW480xenograft tumor expression of E-cadherin, and no significantdifference.Conclusions: Ehuang Decoction retention enema significantly inhibit thegrowth of human colorectal cancer SW480xenograft tumor cells, the exactanti-tumor effect, which can effectively improve the body’s anti-tumorability to reduce the lack of SW480xenograft tumor E-cadherin expression andinhibit tumor metastasis. Provide strong theoretical and experimental studyof the basis for its further used in the clinical treatment of gastrointestinalmalignancies.
