Objective: In order to provide the basis for prevention, diagnosis,treatment and prognosis of intrahepatic cholangiocarcinoma, this researchstudied the expression of proliferating cell nuclear antigen (PCNA) and Ki-67in intrahepatic cholangiocarcinoma and hepatolithiasis tissues, and analysedthe relationship between hepatolithiasis and intrahepatic cholangiocarcinoma.Methods: Taking30cases of normal intrahepatic bile duct tissues ascontrol, this research detected the expression of PCNA and Ki-67in25casesof intrahepatic cholangiocarcinoma tissues and20cases of hepatolithiasistissues by immunohistochemical staining methord (SP). The results wereanalysed by SPSSl3.0statistical software.Results: The expression rate of PCNA in intrahepaticcholangiocarcinoma tissues, hepatolithiasis tissues and normal intrahepaticbile duct tissues were92.0%(23/25),65.0%(13/20) and6.7%(2/30)respectively, the difference was significant (P<0.05). The expression rate ofKi-67in intrahepatic cholangiocarcinoma tissues, hepatolithiasis tissues andnormal intrahepatic bile duct tissues were64.0%(16/25),35.0%(7/20), and3.3%(1/30) respectively, there was a statistically significance (P <0.05). ThePCNA LI (0.456±0.132) of intrahepatic cholangiocarcinoma tissues wassignificantly higher than the Ki-67LI (0.163±0.115)(P<0.05); the PCNA LI(0.287±0.142) of hepatolithiasis tissues was higher than the Ki-67LI(0.086±0.135)(P<0.05). The PCNA expression rates (92.0%23/25) werehigher than the Ki-67expression rates (65.0%13/20) in intrahepaticcholangiocarcinoma tissues (P<0.05); the PCNA expression rates (65.0%13/20) were higher than the Ki-67expression rates (35.0%7/20) inhepatolithiasis tissues, there was a statistically significant (P<0.05). ThePCNA expression rates in intrahepatic cholangiocarcinoma well-moderately and poorly differentiated were89.4%(17/19) and100%(6/6), and thedifference was not significant (P>0.05); Ki-67expression rates(100%6/6) inpoorly differentiated intrahepatic cholangiocarcinoma was significantly higherthan in well-moderately (52.6%10/19)(P<0.05). It was correlated that PCNAand Ki-67expression in intrahepatic cholangiocarcinoma and hepatolithiasistissues (P<0.05).Conclusion: PCNA and Ki-67expression rate were increased graduallyin normal intrahepatic bile duct tissues, hepatolithiasis tissues and intrahepaticcholangiocarcinoma tissues, suggesting that hepatolithiasis may be the causeof intrahepatic cholangiocarcinoma. PCNA and Ki-67expression inintrahepatic cholangiocarcinoma and hepatolithiasis tissue were correlated.
