Object:This study investigates the effect of rhizoma pinelliae, Sichuan fritillary, Zhejiangfritillary on the antioxidant activity of Wutou Decoction; verify the scientific of“eighteen incompatible Chinese medicaments” from antioxidant activity point of view.Method:Through FRAP method and DPPH· scavenging method to investigate the in vitroantioxidant activity of Wutou Decoction and Wutou Decoction added with rhizomapinelliae, Sichuan fritillary, Zhejiang fritillary, investigate the effect of these threeopposing herb medicine to the antioxidant activity of Wutou Decoction; moreoverinvestigate the in vitro antioxidant activity of prepared monkshood, preparedmonkshood combine with three opposing herb medicine and formula medicine, theninvestigate the antioxidant activity of mixture of prepared monkshood with opposingherb medicine and formula medicine, verify the theory of opposing herb medicine affectWutou Decoction.Investigate the therapeutic action of Wutou Decoction and Wutou Decoction addedwith rhizoma pinelliae, Sichuan fritillary and Zhejiang fritillary to the rheumatoidarthritis, moreover investigate the enzyme activity of superoxide dismutase (SOD),glutathione peroxidase (GSH-PX) and the content of malecic dialdehyde (MDA) in theserum of mice, investigate the effect of rhizoma pinelliae, Sichuan fritillary, Zhejiangfritillary on the in vivo antioxidant activity of Wutou Decoction. Result:(1)Wutou Decoction added with rhizoma pinelliae, Sichuan fritillary couldpromote the in vitro antioxidant activity of Wutou Decoction, but the promotion willreduce with the increase of opposing herb medicine dose, prepared monkshood combinewith rhizoma pinelliae, Sichuan fritillary will inhibited the in vitro antioxidant activity;Wutou Decoction added with Zhejiang fritillary could promote the in vitro antioxidantactivity, the promotion to the total antioxidant activity will enhance with the increase ofZhejiang fritillary dose, but the promotion to the DPPH· scavenging assay have norelation to the dose of Zhejiang fritillary, prepared monkshood combine with Zhejiangfritillary could inhibit the total antioxidant activity, but the inhibition will eliminatewith the increase of the Zhejiang fritillary dose, prepared monkshood combine withZhejiang fritillary could inhibit the DPPH· scavenging assay and have no relation to theZhejiang fritillary dose; prepared monkshood combine with formula medicine andprepared monkshood mixture with opposing herb medicine,formula medicine have noinhibition to the in vitro antioxidant activity.(2)Wutou Decoction and Wutou Decoction added with rhizoma pinelliae, Sichuanfritillary, and Zhejiang fritillary are all have therapeutic action to rheumatoid arthritis,and the therapeutic action have related to the antioxidant, but the mice dose with WutouDecoction added with rhizoma pinelliae, Sichuan fritillary, and Zhejiang fritillary havea low level of enzyme activity of superoxide dismutase (SOD), glutathione peroxidase(GSH-PX) than the mice dose with Wutou Decoction, The of the mice dose with WutouDecoction added with rhizoma pinelliae, Sichuan fritillary, and Zhejiang fritillary havea high level of content of malecic dialdehyde (MDA) than the mice dose with WutouDecoction. Conclusion:Rhizoma pinelliae and Sichuan fritillary will inhibit the in vitro antioxidant activityof Wutou Decoction and the inhabitation is due to the combination of preparedmonkshood with them, Zhejiang fritillary will promote the in vitro antioxidant activity,but prepared monkshood combines with Zhejiang fritillary will inhibit the in vitroantioxidant activity yet; rhizoma pinelliae, Sichuan fritillary and Zhejiang fritillarycombine with Wutou Decoction will also have therapeutic action to the rheumatoidarthritis, but these three opposing herb medicine have inhabitation on the in vivoantioxidant activity on the Wutou Decoction.
