Objective:To study the expression of NKD1and P-catenin in colorectal normal mucosa, colorectal adenoma and colorectal adenocarcinoma, analysis their correlation, and investigate their roles in the development of colorectal cancer and assessment of prognosis.Methods:1)Expression of NKD1and β-catenin in18cases of adjacent normal colorectal mucosa,20cases of adenomas and97cases of colorectal adenocarcinoma was evaluated by immunhistochemistry.2) Expression of NKD1mRNA and P-catenin mRNA in10cases of adjacent normal colorectal mucosa and10cases of colorectal adenocarcinoma was evaluated by RT-PCR.Results:1) Positive expression rate of NKD1in adjacent normal colorectal mucosa, adenomas and colorectal adenocarcinoma was gradually reduce(P<0.05). Ectopic expression of P-catenin in colorectal normal mucosa, colorectal adenoma and colorectal adenocarcinoma was gradually rise(P<0.05).2) The expression of NKD1in colorectal adenocarcinoma to be associated with the depth,degree of differentiation, Dukes stages and lymph node metastasis (P<0.05),but not correlate with the patients’age,gender,position and distant metastasis (P>0.05); Ectopic expression rate of P-catenin in colorectal adenocarcinoma correlated with the depth,degree of differentiation, Dukes stages and lymph node metastasis (P<0.05), but not correlate with the patients age,gender,position and distant metastasis (P>0.05).3) The expression of NKD1mRNA in colorectal adenocarcinoma was significantly higher than that in colorectal normal mucosa. The expression of p-catenin mRNA in colorectal adenocarcinoma was similar with that in colorectal normal mucosa.4) Expression of NKD1and ectopic expression of P-catenin were negatively correlated (rs=-0.691, P<0.01).Conclusion:1) Positive expression rate of NKD1in adjacent normal colorectal mucosa, adenomas and colorectal adenocarcinoma was gradually reduce, and to be associated with the depth,degree of differentiation, Dukes stages and lymph node metastasis, NKD1as Wnt signaling pathway inhibitor, plays an important role in inhibiting colorectal tumor development.2) Ectopic expression of P-catenin in colorectal normal mucosa, colorectal adenoma and colorectal adenocarcinoma was gradually rise, and correlated with the depth, degree of differentiation, Dukes stages and lymph node metastasis, suggesting that as key genes of the Wnt signaling pathway, β-catenin plays an important role in invasion and metastasis of colorectal cancer.3) NKD1mRNA express in colorectal cancer tissues was significantly higher than that in the control group of non-cancerous, inconsistent with its protein expression, suggesting that NKD1may have more complex mechanisms have yet to be more in-depth research in colorectal.4) Expression of NKD1and ectopic expression of β-catenin were negatively correlated, combined detection of both may provide some help in diagnosis, treatment and assessment of prognosis of colorectal cancer.
