Objective: To explore the expressions and the clinical pathologicalfeatures of JC virus early region gene encoding product large tumor antigen(T-Ag) and β-catenin proteins in the occurrence and development of colorectaladenocarcinoma, and to study the correlation between the two genes.Methods: The expressions of JCV T-Ag、β-catenin were detected by RT-PCRand immunohistochemistry in65human colorectal cancr tissue and20normalcolorectal tissue with RT-PCR and immunohistochemistry, and to analyze thecorrelation between the two genes and clinical significance combined with theThe sex of the patients,tumor size, location lymph metastasis, differentiationdegree and Dukes staging.Results:The positive expression rate of JCV T-Ag in65cases of colorectalcancer tissues was38.4%(25/65),and there was no expression in20cases ofnormal colorectal mucosa. There was no significant differerce in differentiationdepth of infiltration of colorectal adenocarcinoma a (P>0.05),and there wasopposite in lymph nodes metastasis of colorectal adenocarcinoma a(P<0.05).The positive expression rate of β-catenin proteins in65cases ofcolorectal adenocarcinoma was47.7%(31/65). There was no significantdiffererce in differentiation depth of infiltration and metastatic lymph nodemetastasis of colorectal adenocarcinoma (P<0.05). The expressions of JCVT-Ag and β-catenin proteins was negatively correlated in colorectaladenocarcinoma (P<0.01).To detect the expressions of JCV T-Ag、β-catenin by RT-PCR in65 cases of colorectal cancer tissues and20cases of normal colorectal mucosa,andanalyse their correlation between the two genes and clinical significance. theexpressions of JCV T-Ag、β-catenin in normal intestinal mucosa has beenpublished.The positive expressions of JCV T-Ag in65cases of colorectaladenocarcinoma a were66.15%(43/65),and β-catenin was63.07%(41/65). Theabnormal expressions of β-catenin proteins in65cases of colorectaladenocarcinoma were63.07%(41/65). There was significantly higher than inhealthy control (P<0.05).Conclusions:1. expressions of JCV T-Ag in the early colorectal cancer has an importantrole in the formation process; there was no correlation with age,tumor size, thetumor infiltration depth, the type of histopathology, location lymph metastasisand Dukes staging.2. There were correlations with age,tumor size, the tumor infiltration depth, thetype of histopathology, location lymph metastasis and Dukes staging,soβ-catenin might play an important role in the occurrence, progression andprognosis of colorectal caner.3. The expressions of JCV T-Ag and β-catenin proteins were negativelycorrelated in colorectal adenocarcinoma.we can conclude JCV T-Ag inhibitβ-catenin signaling pathway to promote the formation of tumors.
