| Objective:To analyze the significance of PAR-2,β-catenin and COX-2 in the carcinogenesis and progression of gastic adenocarcinoma,to study the experimental foundation of the prognosis evaluation.Methods:Immunohistochemical SP method was used to determine the expression of PAR-2,β-catenin and COX-2 in 63 cases of gastric adenocarcinoma,19 cases of low-grade intraepithelial neoplasia and 20 cases of normal gastric mucosal tissues,the relations between the expression of them and clinicopathological features and survival rate,and the correlation between PAR-2,β-catenin and COX-2 was analyzed.Results:(1) The positive rates of PAR-2 in gastric adenocarcinoma,in low grade intraepithelial neoplasia and in normal gastric mucosal tissues were 47.6%,21.1%,10%,the abnormal expression rates ofβ-catenin in gastric adenocarcinoma,in low grade intraepithelial neoplasia and in normal gastric mucosal tissues were 55.6%,31.6%,0%,the positive rates of COX-2 in gastric adenocarcinoma,in low grade intraepithelial neoplasia and in normal gastric mucosal tissues were 68.3% 42.1% 30%. (2) The expression of PAR-2 in carcinoma tissue was closely correlated with depth of infiltration (P<0.05),but not with age,gender,differentiation degree,lymph node metastasis and TNM stages;the abnormal expression ofβ-catenin was closely correlated with differentiation degree,depth of infiltration,lymph node metastasis and TNM stages (P<0.05),but not with age and gender;the expression of COX-2 was closely correlated with depth of infiltration,lymph node metastasis and TNM stages,but not with age, gender and differentiation degree (P< 0.05). (3) According to the method of Spearman correlation analysis,the positive expression of PAR-2 and COX-2, the positive expression of PAR-2 and the abnormal expression ofβ-catenin, the abnormal expressionβ-catenin and the positive expression of COX-2 (P<0.05) were related posotively in gastric adenocarcinoma.(4)According to the method of Kaplan-Meier,the difference between the positive expression group of PAR-2,COX-2 and the negative group are of statistical significance in gastric adenocarcinoma,the difference between the abnormal expression group ofβ-catenin and the normal group are of statistical significance in gastric adenocarcinoma.Conclusions:The abnormal expression ofβ-catenin and the positive expression COX-2 was closely correlated with the carcinogenesis and progression of gastic carcinoma,and they may be valuable prognosic makers. The positive expression of PAR-2 was correlated with the progression of gastic carcinoma,and it may have some reference to value prognosis. |
