| 1.Background and PurposeColorectal cancer is a common malignant tumor, every year, over one million people are diagnosed with colorectal cancer in the world, the morbidity and mortality rates are in the forefront of all malignant tumors and there is an increasing trend. Invasion and metastasis are the important malignant characteristics and are often the main cause to lead patients death. At present,the traditional treatment is not satisfactory.Tumor cell biology which can reveal the mechanism of carcinogenesis has become an issue of universal concern. Transforming growth factor beta1(TGF-β1) is a polypeptide cytokine with a wide functions, playing an important role in the proliferation, differentiation and modulate cell migration. The occurrence and development of many tumors have a close relationship with TGF-β1. VEGF has been found to be the most important angiogenesis factor which can induce neovascularization and increase vascular permeability, eventually leading to the proliferation,invasion and metastasis of tumor lesions.This study through detecting the expression of the two genetic products in colorectal cancer to explore the intrinsic relationship between them and their clinical significance.2.MethodsUse immunohistochemical method to detect the expression of TGF-β1and VEGF in88cases of colorectal cancer tissues and40cases of normal tissues adjacent to carcinoma, and analyze the correlation among indicators from the patient’s age,gender, tumor size, pathologic stage, lymph node metastasis and Dukes stage. 3.Statistical analysisUse X2to test the expression of TGF-p1, VEGF and comparison with pathological parameters. Use spearman rank to inspect the correlation of expression of TGF beta1,VEGF in colorectal cancer tissues. Use Kaplan Meier to describe the survival curve and survival rate, Use the Log-Rank sequence to estimate survival rate difference between the two groups.Statistical software SPSS18.0was used for data analysis. P<0.05presents significant difference.4.Results1) The positive ratio of TGF-β1, VEGF in colorectal cancer was78.41%and72.73%, significantly higher than that in the control group (30%and20%, respectively), in comparison,P<0.05.2) The positive expression rate of TGF-β1,VEGF was associated with colorectal cancer’s clinical stage, lymph node metastasis (P<0.05), while had no significant difference in sex, age, the size of tumor and histological grade (P>0.05).3) The expression of TGF-β1, VEGF in colorectal cancer tissues was positively correlated (p<0.01)4) The analysis of single factorrevealed that the expression of TGF-β1, VEGF was significantly related with the prognosis of patients.(p<0.01)(p<0.01)5.ConclusionThe expression of TGF-β1and VEGF is closely related to colorectal cancer’s invasion,metastasis and prognosis. |
