| Based on a large number of literature, the compounds like naphthoquinones,flavonoids,polyphenols,terpenoids from Juglandaceae, with important pharmacologicalaction of anticancer, bacteriostasis, allelopathy and insecticidal activity appeared marketprospect outstanding. Raw material, Carya cathayensis Sarg. epicarps, are rich andabandoned. Deep research had done to the identification of active substances from Caryacathayensis Sarg.epicarps, and purification of a kind monomericcompound.Specific content as follows:①Through chemical qualitative detection, we have undertaken a systematicinvestigation on the chemical constituents of Carya cathayensis Sarg.epicarps. And thechemical constituents such as organic acids, saponins, anthraquinones, flavonoids, lactones,coumarins, cardiac glycosides, alkaloids, terpenoids and steriods were identified.â‘¡Isolation and purification factors to the content maximum of chloroform extractionwere studied. Firstly,got rid of most part of foreign matters by preparative TLC. On thebasis of single-factor test which determined the type and upper/lower limit of organicsolvents. Quadratic General Rotary Design was first used to get the solvents with volumeratio. Then pure product isolated through C18reverse phase silica gel columnchromatography was the major peak detected by HPLC. Collected crude productssubsection eluent and combined with the flow of above95%purity. The obtained regiolonecould be used as drug standard product for analysis test or pharmacology and toxicologyresearch which the relative content up to98.1%.â‘¢UV, HPLC, EI-TOF-MS(70eV),1H NMR and13C NMR characterization factorsprovide theoretical parameters to rapid detection dispense with standard product in future.Studies showed that maximum absorption wavelengh at260nm and340nm, the molecularweight178,the double bond equivalents6,the characteristic ion peaks at m/z121(97%)ã€m/z132(20.5%)〠m/z160(15%)〠m/z122(37%)〠m/z150(27%)〠m/z149(15.6%).Furthermore, the work studied the mass spectrum characterization of regiolone andspeculated on the fragmentation pathways.â‘£The conditions to hot-reflux extraction of regiolone were studied. To ensure thecontrol of quality in production process, regiolone was determined by HPLC andmethodology was also inspected. On the basis of single factor experimcent, the resultsindicated that95%ethanol was the preferred solvent for regiolone extraction and extracttwice enough. The optimum conditions of orthogonal experiment were as follows:temperature80℃, solid/liquid ratio1:10, extracting time60min. The extraction technologyfor regiolone is scientific, rational and can be used in industrial production. |
