In order to find the optimum extraction and purification techniques and its cytotoxicity, we study on the alkaloids from the peels of Carya cathayensis Sarg. to provide a scientific basis for the further exploitation and utilization.Traditional immersion extraction method was compared with ultrasonic-assisted extraction method and microwave-assisted extraction method. The optimum extracting proceduce was selected with the total alkaloids from the peels of Carya. cathavensis Sarg. by orthogonal test design. The results showed that the microwave-assisted extraction method is superior than traditional immersion extraction method and ultrasonic-assisted extraction method. The extraction conditions was ultimately determined:60% ethanol, microwave radiant intensity 520W, extracting temp.30℃,microwave of extraction 7min after soaked for 24 hours at room temp, and the yield of total alkaloids is 0.94%.Separation and purification of total alkaloids from the peels of Carya. cathayensis Sarg. by macroporous adsorption resin. Three resins(D101,NKA-9,AB-8)were screened by static test. The results show that the resin of NKA-9 has the best overall performance. The purification process parameters were further studied by single factor experiments and orthogonal experiments, including three factors:sample solution pH,eluent concentration and eluent consumption. The method for purification was studied with the total alkaloids purification multiple and solid ratio as indexes.Analysis results by range analysis and variance analysis. The optimum conditions was:pH value of sample is 7.0; elute total alkaloids part with 3BV 50% alconol after eluting with 2BV distilled water and 2BV 30% alcohol. The purification multiple was 3.173,the solid ratio was 34.78%.The cytotoxicity activity of the extract was evaluated by using methods of MTT with cell growth inhibition rate as index.The study showed that the extract significantly inhibited the growth of the cell of HepG-2,K562 and Bcap-37. IC50 were 86.6ug/ml,293.4ug/ml,152.6ug/ml.
