The Research Of Partial Mechanism Of Yi Qi Yang Xue Decoction In Treating Osteoarthritis

Objective:To explore the security and the influence of Yi Qi Yang Xue decoction in IL-1β, TNF-α, apoptosis and cartilaginous tissue.Methods:40Japanese Large Ear rabbits randomly divided into4groups, the sham group, the KOA model group, the positive group (Shuang cu rui ying) and the Yi Qi Yang Xue decoction group. All the experimental animals had free diet for one week before the experiment. The KOA model group, the positive group (Shuang cu rui ying) and the Yi Qi Yang Xue decoction group were manufactured into OA model in the right knee by means of Hulth method. The sham group were only opened the articular cavity.4weeks after the operation, the KOA model group and the sham group were drenched by normal saline while the Yi Qi Yang Xue decoction group and the positive group(Shuang cu rui ying) were drenched by liquids made for each group once a day, for6weeks.10weeks later, the serum and knees were collected for the rest of the research, such as the content of IL-1β, TNF-α,hematoxylin-eosin staining and TUNEL for apoptosis.Results:1. Hematoxylin-eosin staining:Morphology showed that the cartilage structure of the sham group were normal, the KOA model group were changed severely, while the positive group and Yi Qi Yang Xue decoction group changed a little.2.The content of TNF-a in the serum of the KOA model group were higher than the sham group. There were significant difference between them(P <0.05). The content of TNF-a in the serum of the positive group and the Yi Qi Yang Xue decoction group were lower than the KOA model group. There were significant difference between them(P<0.01, P<0.05).3. The content of IL-1β in the serum of the KOA model group were higher than the sham group. There were significant difference between them(P <0.01). The content of IL-1β in the serum of the positive group were lower than the KOA model group. There were significant difference between them(P<0.01). The content of IL-1β in the serum of the Yi Qi Yang Xue decoction group were lower than the KOA model group. There were no significant difference between them(P>0.05).4.The sham group had little apoptotic cells, while the KOA model group had a lot and the Yi Qi Yang Xue decoction group had less than the KOA model group.5. The maximum dosage for mice were195times as much as for human (70kg) Conclusion:1.Rabbit KOA models were successfully established. The sham operation did no injury to the articular cartilage. Yi Qi Yang Xue decoction can protect the articular cartilage.2.Yi Qi Yang Xue decoction can lower the content of TNF-a in the serum to slower the process of cartilage degeneration.3.Yi Qi Yang Xue decoction has little influence on the content of IL-1β in the serum. Needing further research.4.Yi Qi Yang Xue decoction can restrain the apoptosis of the articular cartilage5. It is safe to take Yi Qi Yang Xue decoction for a short time. Needing further research.
