Objectives:With the guidance of Chinese medicine theory, therapeutic effect and function mechanism of Qiwei turmeric Decoction on experimental animal model caused by56°liquor were studied, offering new thought and scientific basis for control acute alcohol poisoning with Chinese medicine.Methods:SPF mice, male,18g±2g, were randomly divided into:control group, model group, RU-21(alcohol metabolism supplement description) group, and Qiwei turmeric Decoction (QWJH) group. In addition to the control group was given normal saline, and the remaining to give56°alcohol toxicity to establish a mice model of acute alcoholism. Before modeling30min preventive give the RU-21, Qiwei turmeric Decoction intervention and control group, model group to give equal volume of saline. Observed in mice drunk, plasma ethanol concentration, serum ALT (alanine aminotransferase) and AST (aspartate amino transferase), the ALP (alkaline phosphatase) activity, measured by ELISA in liver homogenate TNF-a (tumor necrosis factor-a), IL-8(interleukin-8), ADH (alcohol dehydrogenase), the activity of the ALDH (aldehyde dehydrogenase), P450(cytochrome P450), SOD (superoxide dismutase) and MDA (malondialdehyde) to detect brain tissue homogenates of β-EP (β-endorphin), LENK(leucine-enkephalin), optical microscope observation of liver pathology change. The evaluation Qiwei turmeric Decoction a mouse model of acute alcohol intoxication effect, of the hangover, anti-drunk mechanism.Results:1. Qiwei turmeric Decoction group of " Ebriety " time (min) due to alcoholism, RU-21group was significantly prolonged compared with the model group, three groups were304.5±23.27,19.23±6.29、137.1±17.65,P<0.01,at the same time, Qiwei turmeric Decoction group by the " Ebriety time " state to "sober up" state longer than the model group, RU-21group was significantly shortened, three groups were265.50±27.2、661.43±38.56、351.9±35.93,P<0.01.2. Qiwei turmeric Decoction group mice24h survival rate compared with the model group significantly improved the survival rate of the two groups were100%,66%, both more significant difference in x2=11.452,P=0.003,P<0.01.3. Qiwei turmeric Decoction group in each time period (0.5h,1h,1.5h,2h,3h,6h),the plasma ethanol concentration (mg/mL) compared with model group, RU-21group is low, a concentration of31.47±0.67,32.64±0.6,35.77±0.66,37.76±1.11,27.52±1.58,23.81±1.03, P<0.01.4. Qiwei turmeric Decoction group of mice liver homogenates of ADH (U/ml), the ALDH (nmol/ml), cytochrome P450(nmol/ml), SOD (U/ml) activity compared with model group, anditsconcentrationwerel.628±0.174,0.8398±0.06,0.6356±0.25,151.97±7.72, P<0.05.5. Qiwei turmeric Decoction group of mouse brain tissue homogenate, β-EP (U/ml) content LENK (nmol/ml) compared with the model group were1.549±0.25,1.681±0.21, P<0.05.6. Qiwei turmeric Decoction group of mice serum AIT (U/L), AST (U/L), the activity of ALP (U/L) than in model group was significantly lower,75.25±17.77,181±20.52,121±4.96, P<0.05.7. Qiwei turmeric Decoction group of mouse liver tissue homogenate of the TNF-a (ng/1), the level of IL-8(pg/ml), content of MDA (nmol/ml) compared with model group, respectively1.1354±0.225,0.2836±0.02,3.08±1.3, P<0.05.8. Qiwei turmeric Decoction group of mice was significant compared with the model group, liver damage caused by alcoholism reducedχ2=11.719, P=0.001, P <0.01.Conclusion1. Qiwei turmeric Decoction can effectively reduce the alcoholism mouse model of the absorption of alcohol; by increasing the ADH, the ALDH and P450, the activity of SOD way to accelerate the metabolism of alcohol, which significantly extend the lead to the " Ebriety " time due to alcoholism, a significantshorten the mice by the "Ebriety time " state to "awake" state time, significantly increase the mice24h survival rate, thus demonstrating significant "hangover" effect; its hepatoprotective mechanism and reduce the plasma concentration of ethanol; elevated ADH, ALDH, P450, SOD activity and decreased MDA content, speed up alcohol metabolism; reduce β-EP、LENK activity, reduce to xic substances into the brain related.2. Qiwei turmeric Decoction before to reduce significantly lower serum ALT, AST, and ALP activity was significantly alleviate the liver injury caused by acute alcohol intoxication, which showed significant hepatoprotective effect. Its hepatoprotective mechanism and reduction of liver tissue homogenate of TNF-a, IL-8activity, reduce the inflammatory response.
