Objective:This study focused on a hypothesis that the relevance of acupoints and brain has specific metabolism material basis, with the non-acupoint as control, we using ’H-NMR technology and methods of pattern recognition to find the brain metabolism related to"Zusanli""Shangjuxu""Tianshu". to recognite the differences of brain metabolite of acupoints and non-acupoint, and to provide objective basis for explaining the brain metabolism substance basis and target of acupoints and brain,finally we hope it can provide a new research thought for further exploring the relevance of brain and acupoints.Methods:Selecting SD male rats, devided the rats into normal group, model group, acupoint group, non-acupoint group at random. Taking "Zusanli""Shangjuxu""Tianshu"to acupuncture once everyday at the same time, for five days.’H-NMR technique was used to obtain the brain metabolism,the data were normalized, integrated and treated by SIMCA-P10.0software pack to analize the major component (PCA) and the partial leastquares discriminate (PLS-DA) in combination with OSC method, in this way, the corresponding score scattering plot and loading plot were obtained.Result:1. After molding, using benzidine to detect the rat ccult it showed positive, light microscopic and electron microscopic showed intestinal mucosal edema and inflammatory cell infiltration.These prove that the model has typical pathological chang of ulcerative colitis.2. Acupuncturing at"Zusanli""Shangjuxu""Tianshu" can significantly improve the UC symptoms,but acupuncturing at the non-acupoint is not clear.This suggests that acupoint is more effective than the non-acupoint.3. The metabolism research on cerebral cortex of UC ratUsing PCA/OPLS-DA to analysize,we found Leucine/Isoleucine, Valine, Acetate, GABA, Choline, Alanine of model group increased significantly; and Lactate, Glutamate, Phosphocreatine, NAAG, NAA, Myo-Inosital were decreaed.In fat solube metabolites, LDL of the model group was increased, VLDL was decreased.4. comparative study of cerebral cortex metabolite of acupoint group and non-acupoint groupIn water soluble metabolites,Lactate,Glutamate,Phosphocreatine,NAAG,Ribose of acupoint group increased significantly.Valine,Acetate,GABA,Creatine,Asparate of non-acupoint group were decreased.In fat solube metabolites,LDL of acupoint group was decreased,VLDL was increased.Conclusions:1. compared to non-acupoint group, acupoint group had therapeutic effect on UC rat,but non-acupoint group is not obvious.2. The study showed that the rat cerebral cortex metabolite of acupoint group had obvious influence,especially on the water solubility metabolite.It is necessary to further study the key metabolites,such as Lactate, Phosphocreatine, NAAG, LDL, VLDL.The key metabolites of non-acupoint group are GABA and Valine.This showed that the brain metabolites of non-acupoint group are obviously different to acupoint group.
