Aim:To investigate the effect of selecting the acupoints according to meridians treating essential hypertension, High Performance Liquid Chromatography-Multiple Reaction Monitoring (HPLC-MRM) was used to study the target metabolism changes in plasma, and the regulating effect of needling on targeted small molecules, therefore, to provide new clues and biological evidences for the enrichment of mechanism of selecting the acupoints according to meridiansMethods:1.Study on targeted small molecules of EH patients:EH patients were recruited in and24-hour ambulatory blood pressure was evaluated; Based on the EH-closely-related metabolism molecules found in previous studies, HPLC-MRM techniques was used to monitor the changes of target metabolite. By calculating the correlation between the key metabolites and the blood pressure value, the key metabolites were founded and the significances were analyzed.2. Studies on targeted small molecules of Selecting the acupoints according to the meridians:Treatment principle of selecting the acupoints according to the meridians was chosen to treat essential hypertension, similarly, HPLC-MRM techniques was used to compare the amount changes of target metabolites in EH patients plasma before and after treatment, focus was placed on observing the regulating effect of treatment principle on the key metabolites.Results:1. Comparison between EH patients and healthy volunteer, the level of fructose, galactose, tryptophan, tyrosine, oleic acid, sucrose, urea were all significantly increased, however, the level of hexanoic acid was significantly decreased.2. The level of sucrose was positively related to the systolic blood pressure(P<0.05); urea was negatively related to the diastolic blood pressure(P<0.05); galactose was positively related to the diastolic blood pressure (P<0.05). Nevertheless, by the relativity analysis between other key metabolites and blood pressure level, it turned out that there is no obvious relativity between those.3. After treatment, the level of threonine, tryptophan and urea were increased, the level of sucrose and inositol were decreased, and glactose has a downward trending, but it doesn’t have any statistical differences.4. The main changed metabolites after acupuncture treatment are amino acids, saccharides and fatty acids, those metabolites participate in energy metabolism or are intermediate products or end products of energy metabolism.Conclusions:1. There exist the turbulence of metabolites in EH patients, among which, urea, sucrose,and galactose were related to blood pressure value, those metabolites maybe are the key metabolites.2. The treatment principle of Needling according to meridians can regulate the turbulence existing in EH patients in a good way, and with a different intensity for different metabolites. The regulation mainly involves with the small molecules which has a close relationship with energy metabolite. Since energy metabolism is closely related to hypothalamus, we conclude that acupuncture can regulate the function of hypothalamus to promote energy metabolism, decrease the accumulation of energy substance, reduce the oxidative stress response induced by fatty acid, recover the function of vessels, and so on. It can improve the condition of EH patients, furthermore, to lower down the blood pressure.3. Compared with traditional metabolomics technology, taget metabolites research can reflect the different regulating function for treating EH by acupuncture, and it can supply a new technology and direction on study of acupuncture.
