ObjectiveTo investigate the expression of insulin-like growth factor-I(ⅡGF-Ⅱ)andmatrix metalloproteinases-9(MMP-9) in the villi of women with normalearly pregnancy and early spontaneous abortion and diseuss their role in thepathogenesis of early spontaneous abortion.MethodsWe selected20women in early nomal pregnaney and30women in earlyspontaneous abortion that seeked he1ps in the family planning clinic,Tai’ancentral hospital between the march2011and october2011. The villous tissueof the two groups were gottenin the asepsis operations and divided into twoparts、respectively.One part of villous tissue was snap-frozen in refrigerator of-70℃for RNA extraction and was detected the expression of IGF-Ⅱ andMMP-9mRNA by real-time fluorescence quantitative Polymerase ChainReaction (PCR).At the same time,the others was putinto10%formalin andthen embedded them with paraffin.The expression and distribution of IGF-Ⅱand MMP-9protein in the villus of the two groups were observed withtechnique of immunohistochemical staining.Results1.IGF-ⅡmRNA and MMP-9mRNA were both expressed in the villi of the two groups,but they were less significantly in the abortion group than thenormal early pregnaney group. The relative expression of IGF-ⅡmRNA innormal early pregnaney group and abortion group were2.16土0.246,0.632土0.194and MMP-9mRNA in normal early pregnaney group and abortiongroup were abortion group were0.281士0.052,0.152土0.039,the proportionwas signifieantly different(P<0.05).2. Immunohistochemistry analysis tested that the levels of IGF-Ⅱ andMMP-9were both significantly lower in the abortion group than in thenormal early pregnaney group. The mean optical density of two groupswere0.539土0.020,0.271土0.032and0.464土0.061,0.237土0.085. therewas a positive correlation between them and the proportion was signifieantlydifferent(r=0.491,P<0.05).ConelusionThe protein of IGF-Ⅱ and MMP-9participated in the development of inthe villi and their expression were significantly lower which was closelyrelated to early spontaneous abortion, which indicating that IGF-Ⅱ andMMP-9may play importent role in the early spontaneous abortion...
