Objective To compare the different expressions of TGF 1 and its receptors and TNF a in villi and decidua of spontaneous abortion after IVF-ET and those of natural pregnancy and normal early pregnancy, to understand the expressions of TGF 3 1 and its receptors and TNF a in early pregnancy tissues and their effects on early embryo development, to explore the complicated pregnancy process and the mechanism of spontaneous abortion in order to increase the success rate and lower the abortion rate of IVF-ET. That will provide a new approach for studying on the method of biological therapy for non-pregnancy and spontaneous abortion and terminating early pregnancy.Methods By immunohistochemical technique, expressions of TGF 1 and its receptors and TNF a were tested in villi and decidua from 10 cases of spontaneous abortion women after IVF-ET(IVF-ET group) and 20 cases of normal spontaneous abortion women(spontaneous group), and were compared with the villi and decidua from 20 cases of normal early pregnancy women (normal group).Result (1) TGF 1 and its receptors have high expressions in early pregnancy decidua cells, villus glands and interstitial cytoplasm. IVF-ET group, spontaneous group and normal group had the same expression position. Treated by computer photograph analysis system, the average light density of the expressions of TGF 1 in the villi and decidua of IVF-ET group, spontaneous group and normal group were 0.1633 0.7712 & 0.1701 0.8461, 0.1927 0.9854 & 0.2049 0.7594, and 0.2805 0.1188 & 0.2756 0.4832, respectively. The two abortion groups were statistically prominent compared with the normal group (p<0.01). The average light density of the expressions of T 3 RI in the villi and decidua of IVF-ET group, spontaneous group and normal group were 0.1659 + 0.7886 & 0.1751+0.2831, 0.1969 + 0.0061 & 0.2069 + 0.8128, and 0.2702 + 0.7962 & 0.2702 + 0.7004, respectively. The two abortion groups were statistically prominent compared with the normal group (p<0.01). The average light density of the expressions of T RII in the villi and decidua of IVF-ET group, spontaneous group and normal group were 0.1785 0.7589 & 0.1360 0.6263, 0.1509 + 0.9410 & 0.1643 + 0.9597, and 0.3047 + 0.7360 & 0.3035 + 0.0279. Compared with the normal group, the two abortion groups have significant difference (p<0.01). The expression of TGF 1 and its receptors in the early pregnancy tissues of spontaneous aborted women declined.(2) TNF a has expressions in all early pregnancy tissues. The average light desity of the expression of TNF a in villi and decidua of IVF-ET group, spontaneous group and normal group are 0.2740 0.6624 & 0.2599 0.3009, 0.2405 0.9888 & 0.2289 0.6892, and 0.117 0.3117 & 0.1399 0.9254, respectively. Compared with the normal group, the two abortion groups have significant difference (p<0.01). The expression of TNF a in the early pregnancy tissues of spontaneous aborted women rose.Conclusion (1) TGF and its receptors and TNF a adjust reproductive immunuity and effect maternal-fetal interface. They are important trophoblast cell regulators that can form the cytokine environment to progress embryo immune tolerance.(2) Descending of the expression of TGF 3 1 and ascending of the expression of TNF α furtherly confirms that TGF β 1 can impede monocyte from producing TNFα .(3) The mechanism of spontaneous abortion is complicated. This study furtherly confirms that normal pregnancy is a special type of peripheral immue tolerance, while spontaneous abortion is the outcome of the destruction of this kind of tolerance. The abnormal expressions of TGF β 1 and its receptors and TNF α play an important role in spontaneous abortion, they are one of the causes of abortion. |
