PurposeBy the rat the intestinal adhesions animal model to observe the Seprafilmanti-adhesion film role in the prevention of postoperative intestinal adhesion in rats,Seprafilm anti-adhesion films provide a theoretical basis and technical methods in theprevention and treatment of surgical abdominal adhesions.MethodHealthy SD rats170.The Ellis Preparation intestinal adhesion model.Animalswere randomized into four groups, model group (A, n=40) intraperitoneal adhesionssurgery, after surgery without processing; Seprafilm group (B, n=40), intraperitonealadhesions surgery, take Seprafilm anti-adhesion film placed in the wound of cecum;laparoscopic poke holes in the model group (C group, n=40) intraperitonealadhesions surgery, after surgery without processing; the Seprafilm laparoscopic poke holes group (D group, n=40) intraperitoneal adhesions surgery take Seprafilmanti-adhesion film placed in the cecal wound hole and poke card.The rats were killedafter surgery of3,7,14,21days, each batch of each group of10were killed.Visuallygrading rats degree of abdominal adhesions, while observing the wound healing andabdominal infection. Specimens from the most significant damage cecum andabdominal wall adhesions and abdominal wall incision organizations, to observe thepathological changes and the7d observed cecal and the abdominal incisionorganizations Hyp level detection.Results1. Wound healing and abdominal cavity infection:In addition to the fist day after the A group a rat died of anesthetic overdose, after thefirst3day C group a rat died of postoperative infection.,4days after surgery the Agroup of rats died of intestinal obstruction outside.No infection, not yet healed, nointra-abdominal obvious infection postoperative abdominal incision of the rats inother groups after3days surgery. It is visible in the cecum wound of group B andabdominal wall stamp card of Group D at the Seprafilm was transparent gel. The ratshave healing abdominal incision and without the presence of infection in the abdominal cavity after3day,7day,14day,21day.2.Visual observation of the degree of abdominal adhesions:The naked eye cansee adhesions in the part of each group after the surgery of3day. Model group andlaparoscopic poke holes model group are obvious adhesions after7day,14day,21day.Seprafilm reduce postoperative adhesions obviously, the A group and B group wassignificantly different (P <0.05), C group and D group was non-significant difference(P>0.05).3.Organization pathological changes in the light microscope:3day afteroperation: A, C, D group of inflammatory cells infiltrating the cecum whole layercongestion significantly, reddish exudate, fibroblast proliferation, little newborn smallblood vessels damaged cecal pathological change; A, C, D group parietal peritoneumpathological changes similar surface fibrin exudation, banded, large quantities ofneutrophil infiltration, histiocytosis can be seen a small number of new capillaries.7day after operation: Serosal layer was significantly thicker fibrous tissue hyperplasiaactive in A, C, D group, there are amount of fibroblasts and collagen fibers, visiblefreshmen capillary proliferation, and accompanied by a large number of chronicinflammatory cells, the visible portion of necrotic cells; A, C D group of pathological changes similar to the surface of the inflammatory exudate, banded, large quantities ofneutrophil infiltration histiocytosis.14day after operation: Pathological changes of theinjured cecum: A, C, D group were more inflammatory cell infiltration, the visiblepart of the inflammatory cell degeneration and necrosis, fibroblasts arranged in rows,fibrous connective tissue, the proliferation of new blood capillaries; the parietalperitoneal adhesion tissue morphology histological changes: A, C, D groupspathological changes are similar, such as granulation tissue rich and dense, massiveneutrophil infiltration, fibroblasts and capillary proliferation. Pathological changes ofin each group after operation21day and14day are similar. It is a lesser extent lesionsin group B.4. Hyp levels were measuredCecal tissue Hyp level determination: determination of protein content and the levelHyp in adhesion intestinal tissue, the result indicate that adhesion intestinal wall Hyplevels in group B were significantly lower in group A (P <0.05), C, D group adhesionintestinal wall Hyp level was no significant difference (P>0.05), DescriptionSeprafilm can a significant reduction in adhesion intestinal wall Hyp content, inhibitthe proliferation of fibrous connective tissue, reduce the degree of intestinal adhesion intestinal adhesion model.ConclusionSeprafilm anti-adhesion film can significantly reduce intestinal adhesionintestinal adhesion model incidence and degree of adhesion, improve adhesion site ofpathological damage. It is no significant effect on the healing of abdominal incision.The Seprafilm anti-blocking film inhibit hyperplasia of fibrous connective tissue, andto reduce the content of the Hyp in cecum.
