The Research On The Preparation Of The Peritoneal Mesothelium Cell Membrane And Preventing The Harmful Peritoneal Adhesion

Objective:There are many reasons of the peritoneal adhesion.In peritoneal adhesion some researchs reveal that trauma,inflammation,foreign body are mostly factors. This experiement expects to observe a dynamic process of peritoneal adhesion, caused by peritoneum damage and understand the cell mechanism about the peritoneal adhesion,especially the role of the mesothelial cell,and in order to ensure the quantity and the stability,we made the the hemaleucin membrane.In this thesis we did the research on cultivante mesothelial cell on the hemaleucin membrane to unite the hemaleucin membrane with mesothelial cell.we will do the research about transplanting the membrane of hemaleucin-mesothelial cell to confirm its role in preventing and curing the traumatic peritoneal adhesion.Methods:In part 1,one group was the trauma group,another group is normal group,to put to death and observe their status of peritoneal adhesion.The adhesion tissues were taken to graded by the standards and to dye in HE.The datas from measuring were analysed with Ridit analysis.In part 2,we used membrane digestion method and detached the epiploon mesothelial cell from the rats' body,to have a serial cultivation,observe the cell morphous,calculate cell purity and identify the cell by immunity class.In part 3,we did this thesis about the hemaleucin membrane's preparation,its physics performance measure,biocompatibility and degradation.In part 4,we would do the research on cultivante mesothelial cell on the hemaleucin membrane.Conclusion:1. The damage of mesothelial cell and fibroblast proliferation are the basic cytology mechanism of peritoneal adhesion,The consecutive two parts of peritoneum damage is the space foundation.2.we use membrane digestion method and detach the epiploon mesothelial cell from the rats' body successfully,to have a serial cultivation.3.Hemaleucin membrane has good biocompatibility and degradation. 4.we can find that the origin mesothelial cell form the epiploica can grow on the hemaleucin membrane,so we can get the membrane of hemaleucin-mesothelial cell. Mesothelial cell can grow on the hemaleucin membrane normally.5.we find it is helpful to transplant the membrane of hemaleucin-mesothelial cell to preventing and curing the traumatic peritoneal adhesion.It can increase the number of the mesothelial cell,decrease the exudation and prevent the direct contact of the consecutive two parts of peritoneum damage.Transplanting the membrane of hemaleucin-mesothelial cell can decrease rats' peritoneal adhesion obviously.
