Panax ginseng is a world famous precious Chinese herbal medicine andit is known as the “king of herbs” in ancient China. It can both reinforceQi and promote production of body fluid, which means to build uppeople’s health. In this paper, three methods for extraction ofginsenosides in Panax ginseng was studied; inspecting the optimumconditions for the separation of ginsenosides and oligosaccharides inginseng fibrous root with macroporous resin; the quality standards ofginsenosides in ginseng fibrous root was drafted.1. The effect of three extraction methods for the ginsenosides inPanax ginseng: In this study, through orthogonal experiment wedetermined that the best technological formulation of Flash-typeextraction is: three times extraction; ratio of solid to liquid is1:20; thetime for every extraction is3min. verification test showed this method isfeasible. In addition, we analyze the effect of using three methods in theextraction ratio of the ginsenosides in Panax ginseng by ways ofultraviolet-visible spectrophotometry, high-performance liquidchromatography (HPLC) and electrospray ionization mass spectrometry(ESI-MS). The results showed the extraction ratio of ginsenosides inPanax ginseng via Flash-type extraction, circumfluence and ultrasonic isrespectively1.8%,1.6%,1.4%. ESI-MS semi-quantitative analysisshowed that the relative intensity of panaxdiol type ginsenosides andpanaxtriol type ginsenosides in the circumfluence, ultrasonic andFlash-type extraction is respectively771.52%and323.44%;705.93%and405.50%;791.82%and316.24%. The results showed that radios underthree kinds of methods on extracting are similar. But the time they varygreatly, circumfluence cost6h, ultrasonic cost30min and Flash-typeextraction cost only6mins which means that Flash-type extraction is the most efficient.2. Separation and purification of ginsenosides and oligosaccharidesin ginseng fibrous root by D101macroporous resin: We established thebest technological formulation of circumfluence extraction according tothe orthogonal experiment:10volumes of80%ethanol extracted3times,1hour each time; solution concentrated from the method above (0.2g/ml)was purified by D101macroporous resin and we come to the conclusionthat the optimum condition for separation and purification ofginsenosides and oligosaccharides in ginseng fibrous root is: themaximum sample volume is7/6times the column volume; the volume ofwater elution is4times the column volume; eluted with70%ethanol; theelution volume is8times of column volume.3. Research on quality standard of ginseng fibrous root: In thisthesis, we studied the precision, Repeatability, Stability and the recoveriesby means of UV-visible spectrophotometer and HPLC, and establishedquality standards of extraction from ginseng fibrous root. A practicablestandard for the quality control of medicinal herbs was provided.
