[OBJECTIVE]The micro smashing technology is a new technology which develops in nearly20years. It is draws support the special mechanical power, jet flow ability or physics,the chemical action. After microing of the ordinary material, material the surface and the contact surface has the specific change,the traditional Chinese achieves the cell level smashing, the cell breaking through a wall rate enhancement,thus the promotion traditional Chinese medicine ingredient dissolves. As a new technology, traditional Chinese medicine superfine grinding endowed with traditional Chinese medicine of more high-tech content, effectively promote the Chinese native medicine preparation and raising the level of the development of the cause of Chinese traditional medicine. The main chemical compositions of Realgar is As4S4or As2S2, the main elements are arsenic and sulfur, among them the weight of arsenic and sulfur percentage of1.27:1. This thesis is to comparatively study on the dissolution quantity and acute toxicity of traditional powder and ultra-micro powder of Realgar, and study on the stability of ultra-micro powder of Realgar. The results supply the scientific basis on the preparation and clinic application of the ultra-micro powder of Realgar.[METHODS]Ⅰ. Comparative study of powder characterization:The appearance, surface area and particle size are studied. The change of the moisture absorption and fluidity of ultra-micro powder were studied by determining the speed of moisture absorption and angleofrepose. Ⅱ. The dissolution research of effective constituents:Choose arsenic as the effective constituents of the Realgar, determined by atomic fluorescence spectrophotometry according to GB/T5009.11-2003, comparatively study on the vitro dissolution velocity and quantity. Ⅲ. The research on the stability:Study the powder content and particle size of water and Realgar in9months.Ⅳ. The research on the acute toxicity:the acute toxicity of the ultra-micro and normal powder of Realgar was researched by MTD (maximal tolerance dose) of ig the normal and ultra-micro powder powders of Realgar on rabbits.[RESULTS]Ⅰ. The particle size and color of ultra-micro realgar powder are significant changed. The aerated density declined from0.4196g/cm3to0.3960g/cm3and the moisture absorption increased from10.35%to 11.93%.Ⅱ. The study on the dissolution of chemical components in realgar suggests that the quantity of the main components of the ultra-micro powder were enhanced. The quantity of the extract of arsenic in ultra-micro powder is2.1142mg/g respectively, which incressed than normal powder is0.757mg/g. The dissolution paramenters as follow:The T0.5and Td of arsenic decreased from113.61min、176.22min to46.38min、79.16min respectively. That is to say, there is a obvious increase on dissolution rate of components after the ultra-micro powder preparation. Ⅲ. The research on the stability suggests that ultra-micro powder storage in the bag9months, the content of water increased form4.232%to4.975%, and the content of arsenic was92.34%after9months, the powder particle size did not change. IV. The research on the acute toxicity:The acute toxicity experiment makes clear that the toxicity of the ultra-micro powder and normal powder of realgar is large relatively. The MTD of realgar normal powder in mouse is22.751g/kg, while the MTD of ultra-micro powder is11.706g/kg. So the ultra-micro powder can increase the acute toxicity of realgar.[CONCLUSSION]The particle size and color of ultra-micro realgar powder are significant changed, the liquidity is decreased and hygroscopic is increased. The total dissolution and acute toxicity of ultra-micro powder increased comparing with those of normal powder, and the ultra-micro powder storage in the sealed bag is stable. The results provide the basis for the further study on the application, which is cheerful prospect about saving natural resources of Chinese herbal medicine.
