Objective:To study the effects of ultra-micro Qiweibaizhusan on metabolism diversity of intestinal microbes, and to provide the experimental evidence for the curative effect mechanism of ultra-micro Qiweibaizhusan.Methods:This experiment had studied some factors which could influence the quality,quantity and bioactivity of gut microbes extracted,like concentration and volume of NaCl solution, shakened time,speed of centrifuge,the times of high-speed centrifuged.To establish an effective extraction method of intestinal microflora,which would be used to study the intestinal microflora based on culture.The mice model with dysbacteriosis were treated with antibiotics mixture composed of Cefradine Capsules and Gentamycin Sulfate injection by intragastric administration,and then one treatment group mice were given the traditional decoction of Qiweibaizhusan, the other treatment groups’ mice were given1/2amount of ultra-micro powder Qiweibaishusan,and the normal group and model group mice were given with steriled water by intragastric administration.After three days treatment the intestinal microflora were extracted, and their metabolism were determined by Biolog Microbial Identification System.Results:The intestinal content and0.80%~0.90%of NaCl solution had been blended in conical flask with right amount of glass beads,according to quality volume ratio (g:mL) is not bigger than1:10.They had been vortex shanken30min by120rpm,precipitation had been removed following centrifuged5min by500r/min. Then the supernatant had been tubed to EP tube,and high-speed centrifugal ed once by12000r/mim.The supernatant had been abandoned, adding0.80%~0.90%NaCl solution in the EP tube,had made the microflora homodispersed completely by vortex shaking.At last,the intestinal microflora in mice with extracted completely and high quality had been got.Results showed that all groups of AWCD tended to stable through72h of cultivating,and the normal group> the1/2volume of ultra-micro powder group> the traditional decoction group> the model group.The microbial diversity index including Shannon index,Shannon evenness, Simpson index and Mclntosh index of the normal group were the largest,and they were very significantly differences compared with other groups(P<0.01).The Shannon index,Simpson index of the1/2volume of ultra-micro powder group and the Shannon evenness,Mclntosh index of the traditional decoction group were in the second.The results of clustering analysis and principal components analysis suggested that bacteriostasis of antibiotics was obvious,and some metabolism ways of intestinal microbial in mice were influenced.The metabolism diversity of intestinal microbes in mice were gradually regained balance by the regulatory functions of1/2volume of ultra-micro powder and traditional decoction of Qiweibaishusan.There were similarities and differences in regulation roles of the two species decoction,and the regulation role of1/2volume of ultra-micro powder was more notable than traditional decoction.Conclusion:An effective extraction method of intestinal microflora was established,which had extracted enough many microorganisms in intestine,and kept them alive.The regulation role to metabolism diversity of intestinal microbes in mice of1/2volume of ultra-micro Qiweibaizhusan was more notable than traditional decoction,which had a good treatment effect on diarrhea with intestinal dysbacteriosis.
