Objective:There isn’t definite provision for the ultra-fine powder of The Chinese medicine power in our country. But quality and effective components of the power as important factors in the Chinese medicine Efficacy influence.It must be that control the power quality as it source, and form a complete Control Architecture, therefore, the key technical points are as followed:Preparation Technology and Establishment of the Quality standard.However, different diameter of the Chinese medicine power Possible relationship between all these problems were urgent. This thesis choose radix saoshnikoviae as the research subject, discussed the technical preparation and the standard of quality of the radix saoshnikoviae powder. The optimization range of the particle diameter is confirmed by tests of different kinds of particle diameter dissolution in invitro.Through the dissolution behavior tests,the most effective abstraction technique is obtained, and provided the scientific basis for the future development of the Chinese medicine powder.Methods:1.The research of the technical preparation for the saposhnikoviae radix. Using D90 as the index, the relationship between crushing result and some influencing factors such as crashing time, temperature, water content, filling rate of medium are studied by applying homogeneous design.2.The research of the physical characteristics of the radix saoshnikoviae: the surface area and the powder density are studied. By the determine of the angle of repose, and the rate and percentage of the moisture absorption, the variation of the mobility and the moisture absorption are examined.3.The research of the quality standard of the saposhnikoviae radix:in order to establish the quality control system, researches are done from different aspects,such as microcharacteristic,TLC identify, heavy metals and as salt, content measurements,and stability.4.The research of the effective component’s dissolution behavior: according to macrotin glyocoside,macrotin,5-O-methylvysemaishamy glyocoside,haikayaphenol glyocoside,examine the elements which influent the abstraction of the radix saoshnikoviae powder, such as time, temperature and method of the abstraction.5.The research of the dissolution in vitro:by researching the volume and the rate of the dissolution in vitro of varied particle diameter of the powder, and the dynamics analysis, to examine the changing feature and pattern of above four effective components.6.Ebacteriostasis activity about saposhnikoviae radix:Using MIC as the index,the relationship between particle size and bacteriostasis activity is studied by applying tube dilution method.Results:1.The optimization technical condition of ultra-fine powder crushing of radix saoshnikoviae is determined:medium filling rate is 60~70%,water content is 4~8%,temperature is 0~10癈,crushing time is 8-16 minutes, material mass is 200g.2.The study of physical and chemical characteristic indicated that the angle of repose of the powder, surface area,the equilibrium moisture absorption and the size of particle diameter are in inverse ratio;the density and the size of particle diameter are in direct ratio.3.The methodology of the chromone against radix saoshnikoviae is established.4.The research of the dissolution behavior indicated that the best parameter for radix saoshnikoviae powder’s abstraction using water is soaking for 2hours at 80℃.5.The study on the dissolution of chemical components in vitro indicates that the quantity of the extract and rate of dissolution speed of the ultra-fine powder preparation being enhanced, compared to coarse powder and fine powder.6.The study of bacteriostasis activity reveal that MIC of ultra-fine powder is smaller than those of fine powder.Conclusions:With changed appearance,the microcharactor, the moisture absorption and the fluidity, the micron powder altered in many physical features,compared to fine powder.meanwhile, the effective component remain stable.Besides,according to content measurement and the result of dissolution, micron powder increased dramatically. From all above, the establishment of technique and quality standard against saposhnikoviae radix provided the scientific direction for the development of Chinese medicine powder. |
