Objective This experiment established a polycystic ovarian syndrome (PCOS) modelof rat, which studied the expression of annexin Ⅰa nd Ⅳ in follicular granulosa cells andclarified molecular biology mechanism of follicular growing,differentiation, apoptosis andatresia in PCOS. Methods Progestogen jointed human chorionic gonadal hormone(HCG)constructed PCOS model by inducing youthful female SD rat. The expression of annexinⅠa nd Ⅳ was detected by immunohistochemical method,and the quantitative analysis wasperformed by the gray value in model and control group.Results (1) The behavior and signwas no obviously different in model group and control group.The quality of model group[(101.78±5.91) g] was higher than control group [(98.37±7.64) g],with no difference(P=0.058).The ovarian weight of model group [(0.049±0.008) g] was obviously higher thancontrol group [(0.026±0.008) g], and ovarian volume of model group [(48.77±12.82)mL]was obviously higher than control group [(25.35±8.91)mL], with significant statisticallydifference (P=0.00).(2)The serum level of P in model group (19.85±18.32)nmol/L washigher than that in control group (9.62±3.82)nmol/L,as T in model group (0.35±0.14) nmol/Lhigher than that in control group (0.29±0.04) nmol/L (P<0.05).The serum level of FINS inmodel group (16.92±2.79) IU/L was statistically higher than in control group (12.13±3.1)IU/L,as FBG in model group (4.42±0.24) mmol/l higher than in control group (3.96±0.19)mmol/l (P<0.01). The serum level of FSH in model group (1.09±0.57) mIU/L wasstatistically lower than in control group (2.01±0.37) mIU/L (P<0.01).The ratio of LH/FSH inmodel group (1.46±0.73) was statistically higher than in control group (0.96±0.69)(P<0.01),as HOMA-IR in model group (3.35±0.66) higher than in control group (2.14±0.59)(P<0.05).(3) Hematoxylineosin staining revealed that parts of ovarian follicles expanded cysticly,inwhich oocytes or corona radiata disappeared,and granular cells’ layer became thinner inmodel rats.(4)The annexinⅠand Ⅳexpressed in different classes of preantral follicles in both model and control group.When the diameter of follicle reached to60~120μm,the grayvalue expression of annexinⅠin model group (52.363±0.033) was significant lower than incontrol group [(52.415±0.044),P<0.01], meanwhile the annexin Ⅳ in modelgroup(57.340±0.014) lower than in control group [(57.358±0.015),P<0.01],which hadstatistically significant difference.Conclusion This experiment about PCOS animal modelhad similarly changed in ovarian pathological and serum endocrine with PCOS patients,which was an ideal animal model.PCOS was a disease of multiple cystic folliculars andfollicular atresia as its main pathological changes, while apoptosis of granular cells was thebasic mechanism in follicular atresia.The annexin Ⅰa nd Ⅳexpression evidently increased insinusoid follicle and the granulosa cell of apoptosis raised in model group,which illuminatedannexinⅠ and Ⅳpaticipated in the process of apoptosis in granulosa cell and played animportant role.
