ObjectiveTo detect the expression of AnnexinⅡand P19 gene and protein in cholangiocarcinoma and non-tumor bile duct. And explore their relationship with the development and clinical and pathological features of cholangiocarcinoma.Methods1. Collected the Wax samples of 58 cholangiocarcinomas and 15 non-tumor bile ducts. Detection the expression of AnnexinⅡand P19 protein with immunohistochemical method.The results of experiment were processed with the statistical software SPSS11.5.The comparison of sample rate was processed with chi square test.The Correlation between two factors was processed with pearson correlation.2. Collected 17 cases of freshed cholangiocarcinoma tissues and 9 cases of freshed non-tumor bile ducts tissues.Detection the expression of AnnexinⅡand P19 mRNA with Semi-quantitative RT-PCR method.The results of experiment expressed the form of x±s.The comparison of two groups was processed with t test.3. Detection the expression of AnnexinⅡand P19 mRNA with Western-blot method in 17 cases of freshed cholangiocarcinoma and 9 cases of freshed non-tumor bile ducts.The results of experiment expressed the form of x±s.The comparison of two groups was processed with t test.Results1. The positive rates of AnnexinⅡexpression in 58 cases of cholangiocarcinoma and 15 cases of non-tumor bile ducts with immunohistochemical method were 74.1%(43/58) and 13.3%(2/15), respectively.The positive rates of P19 expression in two tissues were 24.1% (14/58) and 73.3%(11/15),the positive expressions of P19 between the two groups had statistically significant differences. The expressions of AnnexinⅡand P19 have no relation with the gender, age, tumor size and jaundice of cholangiocarcinoma.But they are correlated with the tumor grade, clinical stage and lymph node metastasis of cholangiocarcinoma. There is negative correlation between the expression rate of AnnexinⅡand that of P19 in cholangiocarcinoma.2. The changes of AnnexinⅡand P19 mRNA level in 17 cases of freshed cholangiocarcinoma and 9 cases of freshed non-tumor bile ducts were detected by RT-PCR. The photodensity analysis of gel electrophoresis show that the level of AnnexinⅡmRNA in 17 cases of freshed cholangiocarcinomas group (1.2346±0.1345) was significantly higher than that in 9 cases of freshed non-tumor bile ducts group(0.4518±0.0837).The level of P19mRNA in 17 cases of freshed cholangiocarcinomas group (0.2346±0.0724) was significantly lower than that in 9 cases of freshed non-tumor bile ducts group(0.4626±0.0432).3. The grey value analysis of Western-blot's gel photograph showed that the level of AnnexinⅡprotein expression in 17 cases of freshed cholangiocarcinomas group (0.8128±0.0125) was significantly higner than that in 9 cases of freshed non-tumor bile ducts group(0.3561±0.0171). The level of P19 protein expression in 17 cases of freshed cholangiocarcinomas group(0.3417±0.0132) was significantly lower than that in 9 cases of freshed non-tumor bile ducts group(0.7651±0.1021).Conclusion1. The expression of AnnexinⅡand P19 in cholangiocarcinoma and non-tumor bile duct were significantly different.The altered expression of the two factors were closely related with cholangiocarcinoma.2. The expressions of AnnexinⅡand P19 are correlated with the tumor grade,clinical stage and lymph node metastasis of cholangiocarcinoma. They may serve as a auxiliary marker for the clinical'process of cholangiocarcinoma.3. There is negative correlation between the expression rate of AnnexinⅡand that of P19 in cholangiocarcinoma.But the related mechanism was needed further confirmed.
