Anovulatory dysfunctional uterine bleeding (ADUB) composes approximately85%inall patients of Dysfunctional uterine bleeding (DUB).The main pathological morphologyof ADUB were the endometrial hyperplasia(EH).However, the apoptotic signalingpathway in EH is unclear. The molecular mechanisms invoved in the progression ofendometrial hyperplasia are unclear, which has hampered the development of an effectiveprevention.Objective: To investigate the modification of signaling pathways related to apoptosisand their roles in the development of endometrial hyperplasia (EH) of rat,and explore thethe related mechanisms of survivin/caspase-3on proliteration and apoptosis in endometrialhyperplasia.Methods:40Sprague-Dawley rats were randomly divided into four groups (n=10);control group(A), model group(B), big dose of Diaomozhibengfang(C), small dose ofDiaomozhibengfang(D),the model of endometrial hyperplasia was induced by the injectionof oestradiol benzoate intramuscularly. Rat in B,C,Dgroup,bilateral ovaries of all rats wereremoved through surgery,3days later;60<mu> g/100g oestradiol benzoate injectionswhich were diluted by high-pressure sterilized peanut oil were intramuscularly injected torat’s once alternate days for4weeks.Then,model of rats with endometrial hyperplasia werebuilt.Later, in group B, normal saline solution was injected into each uterine cavity. Ingroup C,0.3ml big dose of Diaomozhibengfang was injected into each uterine cavity ofthe rat. And in group D,0.3ml small dose of Diaomozhibengfang was injected into eachuterine cavity of the rat.3days after the administration, the rats in all groups weresacrificed. The morphology in uterus was observed under light and the expression levels ofsurvivin and caspase-3were checked by immunohistochemistry and real-time PCR. Results:1.4weeks after the models were induced; the edema and hypertrophy inuteri were observed.The glands in endometrial increased; indented hyperplasia of glandularcells appeared; pseudo-stratified phenomenon occurred. Group C: muscular and serosallayer with infiltration of inflammatory cells, epithelial all necrotic; group D: epithelialmost necrotic, endometrial of rat local inflammation, endometrial part absence, glandsscarce.2. Compared with the control group, the expression of survivin in model groupincreased(P<0.05),while that of caspase-3declined (P<0.05).The proliferated endometriumwas dispelled by Diaomozhibengfang. In the big dose of Diaomozhibengfang and smalldose of Diaomozhibengfang group, the expressions of survivin were reduced and theexpressions of caspase-3were increased (P<0.05).Conclusion:1. We induced the EH model in rats by ovariectomy combining with estrogen.2. Compared with the control group, expression of survivin mRNA in model groupincreased, while that of caspase-3mRNA declined.3. In group C and D, Diaomozhibengfang effects of remove endometrial hyperplasiain rat were obvious. There is no statistically significant in the big dose ofDiaomozhibengfang and the small dose of Diaomozhibengfang.4. The therapy of EH should concentrate on apoptotic signaling pathways: suppressthe expression of survivin, promote the expression of caspase-3, accelerate the apoptoticsignaling pathways and prevent the endometrial hyperplasia.5. Diaomozhibengfang had a targeting function to endometrial hyperplasia rats, whichis expected to be a new drug to treat functional uterine bleeding.
