Objective To investigate the effect of Survivin on MPP~+-induced apoptosis in SH-SY5 Y cells and explore possible mechanisms,provide reference for the study of human PD,reveal the mechanism of PD.Methods SH-SY5 Y cells were assigned to 3 groups.The control group,MPP~+ group,Survivin group.The control group was conventional cultured.The MPP~+ group was induced by 1mM MPP~+.The Survivin group was pretreated by 1.0?g/ml,5.0?g/ml and 10.0?g/ml Survivin for 2 hours before induced by 1mM MPP~+.Each group was cultured for 24 hours.An inverted microscope was used to observe the cell morphology,MTT assay,RT-PCR and Western blot were used to detect related indicators.So that we can assess survivin's effects on the process of apoptosis in SH-SY5 Y cells.Results Observed with inverted microscope,the cell morphology of the control group was present as fusiform or oval.The number of living cells and the cell density of the control group was greater than the MPP~+ group.The light transmission of SH-SY5 Y cells of the control group was much better than the MPP~+ group.Compared with the MPP~+ group,the situation of SH-SY5 Y cells of the Survivin group was better than the MPP~+ group,in a dose-dependent manner.MTT assay showed that 1mM MPP~+ successfully induced the apoptosis of SH-SY5 Y cells,and the cell viability was significantly decreased(P<0.01).However,the cell viability of SH-SY5 Y cells co-cultured with 1mM MPP~+ markedly increased under pretreatmentof Survivin(P<0.05,P<0.01,P<0.01),in a dose-dependent manner.Besides,the results of RT-PCR,Western blot showed that Survivin significantly decreased the expressions of Caspase-3 and Caspase-9 in SH-SY5 Y cells(P<0.05).Conclusion 1mM MPP~+ had the ability to induce the apoptosis of SH-SY5 Y cells.Survivin inhibited the apoptosis of SH-SY5 Y cells induced by MPP~+ in a dose-dependent manner possibly through the Caspase pathways.
