Objective:Coronary heart disease is a common and frequently-occurring disease in the population.It is caused by a combination of factors such as unhealthy lifestyles,environment and genetics.In the development and deterioration of coronary atherosclerotic diseases,genetic and environmental factors are both independent and interactive.The purpose of this study was to explore the relationship between the CD163 gene polymorphism and coronary heart disease,and to explore the relationship between the CD163 gene polymorphism and the plasma lipid levels.Methods:Patients from Affiliated Hospatial of Qingdao University were selected(n=162)and divided into coronary heart disease group(n=87)and control group(n=75).Subjects were on an empty stomach for more than 8 hours without taking lipid-lowering drugs,then were taken venous blood.Total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)were detected by standard blood enzyme colorimetric.Apolipoprotein A(apoA1)and apolipoprotein B(apoB)were measured by immune turbidimetry.Another 3ml of venous blood was placed in the EDTA-K2 anticoagulant tube to extract the whole blood genomic DNA according to the human blood extraction kit operation steps.The extracted DNA was sent to Beijing Nuohe Zhiyuan Bioinformation Technology Co.,Ltd.to amplify the gene target fragment(rs10743940 rs1419980)by polymerase chain reaction(PCR),and directly sequenced for genotyping.Two independent sample t tests were used to compare the serological differences between the subjects of the coronary heart disease group and the control group,such as age,blood lipids,and white blood cells.The chi-square test was used to compare the genotype frequency and allele frequency of the coronary heart disease group and the control group,and the relationship between different genotypes and blood lipids of the coronary heart disease group and the control group was analyzed by single factor variance.Comparison of the relationship between different genotypes and blood lipid in coronary heart disease group and control group was analyzed by one-way ANOVA.Multi-factor Logistic regression was used to analyze the influencing factors of coronary heart disease group.P <0.05 was considered statistically significant.Results:1.The proportion of males and smoking patients in the CHD group was higher thanthat in the control group(P <0.05);the level of triglycerides in the CHD group was higher than that in the control group(P <0.05);in the CHD group,the level of HDL-C was lower than that of the control group with statistical significance(P <0.05).2.The rs1419980 genotype is mainly distributed in AA type in both CHD and control groups.The distribution of AA type in CHD group is less than that in control group,which is statistically significant(P <0.05).The distribution of GG type in CHD group is more than that in control group,which has statistical significance(P <0.05),the AG type distribution of the CHD group was higher than that of the control group,but that was no statistical difference(P> 0.05);the frequency of allele A in the allele distribution of the two groups was higher than the frequency of allele G with statistical significance(P<0.05).The genotype frequencies of the total population examined were AA 54.32%(88/162),AG 33.33%(54/162),GG 12.35%(20/162);allele frequencies A 70.99%(230/324),G 29.01(94/324).The distribution of rs10743940 genotypes in CHD and control group was mainly TT type distribution,while the alleles were mainly T distribution,but there was no statistically significant difference in frequency distribution and allele frequency distribution between the two groups.3.ApoB level of AA and GG genotypes in CHD group at rs1419980 were significantly higher than AG genotype(P <0.05),TC level of GG genotype was higher than AG genotype(P <0.05).There was no statistically significant difference between the genotypes of blood lipid level in the control group.In the control group at rs10743940,the LDL-C level AA genotype was higher than the TT genotype(P <0.05),the TG level TA and AA genotype was higher than the TT genotype(P <0.05).There was no statistically significant difference between the genotypes.4.Variables with statistically significant differences in univariate analysis(P <0.05):gender,smoking,TG,HDL-C,were used to make a multivariate logistic regression analysis and that showed males and triglyceride levels were coronary heart disease Risk factors.Conclusions:1.The rs1419980 locus mainly expresses the AA genotype and A allele in the population,and its expression in the coronary heart disease group is lower than that of the normal population,suggesting that the incidence of coronary heart disease may be related to the low expression of the A allele.Allele A may be a protective genetic factor for coronary heart disease.2.The GG genotype at rs1419980 may be associated with high levels of ApoB and TC in patients with coronary heart disease.The AA genotype at rs10743940 may be related to the high LDL-C and TG levels in the normal population.
