Purpose:To determin the distribution of Vascular endothelial growth factor (VEGF)gene polymorphism in Han and Uyghur Ethnic nationality in south Xinjiang and to study the difference between urolithiasis patients of the two diferent ethnic groups.and impact to urolithiasis illness frequency rate..Methods:Peripheral blood samples,24-hour urine and upper tract stones were collected 200 cases from urolithiasis patients of 200 Han and 200 Uyghur nationality,and in200 healthy Hans and 200 healthy Uyghurs. All of them come from south Xinjiang or Migrant personnel who lived there at least 10 years,urolithiasis symptom attacked two times at least,Confirming the diagnosis by abdominal plain film,intraveneous pyelographm, urinary B-type ultrasoun etc. excluding hyperuricemia, hypercalcinuria, hyperlipemia, diabetes, hypertension,coronary heart disease, gout, hyperparathyroidism,urinary tumour etc. And without blood relationships each other.Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP),gene sequencing and genetic analysis methods were used in urolithiasis patients of 200 Han and 200 Uyghur nationality,and in200 healthy Hans and 200 healthy Uyghurs. Pair comparing the frequency distribution of VEGF genotype and Allele frequency.Results:1. Age ,sex composition,course of disease,BMR without differ in three groups of different genotype(P>0.05).2. The restriction enzyme cutting results of PCR products were found that VEGF gene existed polymorphism by restricted enzyme BST UI digested .VEGF gene had two alleles which were C and D, and TT of VEGF genotype can,t cut by restricted enzyme BST UI, but C of which can.3. Four groups distribution of VEGF genotype were corresponded with Hardy-Weinberg genetic equilibrium after tested (P>0.05). Which indicated that these data had a good representativeness.4. There was a significant difference in the VEGF genotype between the Hans and the Uyghurs insouth Xinjiang(P<0.05),but the VEGF allele frequency there was not(P>0.05)5. There was no a significant difference between the CC of VEGF-460 genotype urolithiasis patients illness frequency rate with the CT,s and TT,s or between the CT,s with TT,s in Han and nor in Uyghur Ethnic nationality (P>0.05).6. Selecting partial VEGF prducts which analyzed by gene sequencing and genetic corresponded with the VEGF genotype which tested by PCR-RFLP electrophoresis.Conclusion:1.A significant differ in the frequency distribution of VEGF genotype but the frequency distribution of allele not in Han and Uyghur Ethnic nationality urolithiasis patients in south Xinjiang.2.A significant differ in the frequency distribution of VEGF genotype but the frequency distribution of allele not between urolithiasis patients with control groups in Han Ethnic nationality in south Xinjiang.3.A significant differ in the frequency distribution of VEGF genotype but the frequency distribution of allele not between urolithiasis patients with control groups in Uyghur Ethnic nationality in south Xinjiang. 4.A significant differ in the frequency distribution of VEGF genotype but the frequency distribution of allele not between healthy Hans and healthy Uyghurs in south Xinjiang.5.A ethnic differ in the VEGF genotype in Han and Uyghur Ethnic nationality in south Xinjiang.6.VEGF-460 gene polymorphism may not markedly coincident with urolithiasis illness frequency rate and nor urolithiasis susceptibility genetic locus in south Xinjiang.
