Background:Breast cancer is one of the most common malignant tumors of women. It is after operation the first reason that the sufferer relapses and dies to infiltration and metastasis. Tumor markers can be used in early diagnosis, therapeutic effectiveness evaluation, prognosis. Judgment and monitoring recurrence and metastasis of malignant tumor. It is of great value to find an ideal tumor marker for early diagnosis of breast cancer, appreciation of the therapeutic effect and monitoring the prognosis. TPS (Tissue polypeptide specific antigen) is a kind of soluble fragment of 18C termini of cytokeratin and a molecular marker of epidermic tumor. ELISA was applied in this research to investigate the serum expression of TPS in patients with breast cancer and investigate their relationships with clinical and pathological characteristics.Objective:To investigate the serum expression of TPS in patients with breast cancer and investigate their relationships with clinical and pathological characteristics.Methods:53 patients with breast cancer,50 cases of benign breast diseases and 50 cases in normal control group. Enzyme Linked Immunosorbent Assay (ELISA) was applied to test TPS concentration. T-test,χ2-test and Pearson, correlation were applied to determine if there are relationships between the expression of TPS and biological behavior.Result:1.Serum TPS concentration is higher in malignant group than in benign and control group, the difference is of statistical significance (P<0.01).But there is no statistical significance between the results of benign breast diseases and normal control; 2.Mutual comparisons made betweenⅠ,ⅡandⅢstages respectively showed that there was no statistical significance betweenⅡandⅠstage (P>0.05),but the results of other mutual comparison are significantly different (P<0.01); 3.In 53 primarily breast cancer patients who have received modified radical mastectomy, Serum TPS concentration declines, the decline was statistical difference (P<0.01); 4. In 53 primarily breast cancer patients, the concentration of TPS was not statistically different regarding the diameter of the mass (2cm).(P<0.01); the concentration of TPS is statistically significantly different between the positive and negative of ER/PR patients, as well as C-erBb-2. Serum concentration of TPS significantly correlates with ER, PR, C-erBb-2 and Ki67. For TPS in regarding of the axillary lymph nodes metastasis, there are also significantly differences whether the number of metastatic lymph nodes.5. The serum levels of 5 tumor markers in breast cancer patients were significantly higher than those in the benign breast disease in the control group (P<0.05). the sensitivity of breast cancer by detecting the serum markers respectively was 56.6%,26.4%,37.75%,30.2%,and 35.8%, and that was 71.7% by TPS plus CA15-3.6. In general, the sensitivity was56.6%, specificity was 94% for TPS. TPS becomes higher to the sensitivity of diagnosis in earlier breast cancer.Conclusion:1. The high serum expression of TPS in breast cancer clue to the both can be used as tumor marker for diagnosis of breast cancer.2. Serum concentration of TPS significantly correlates with axillary metastasis, ER, PR, C-erBb-2 and Ki67, indicating that TPS is probably useful in evaluation and prognosis prediction of breast cancer.3. TPS becomes higher to the sensitivity of diagnosis in earlier breast cancer.4. The serum markers CEA,CA15-3,CA12-5,TPS and VEGF were much more sensitive markers, and the combined detection of CA15-3 and TPS was an effective method in screening breast cancer and their combined detection can significantly improve the sensitivity of breast cancer diagnosis.
