Objectives:In this research, under the guidance of Traditional Chinese Medicine theory,the occurrence and development processes about acute phase of cerebral infarction withgastrointestinal dysfunction were discussed. The intervention effects on acute phase ofcerebral infarction with gastrointestinal dysfunction by the Tongqiao Huoxue Decoctionwere observed. The results showed that the Tongqiao Huoxue Decoction had protectiveeffect on the brain and gastrointestinal injury which induced by cerebralischemia-reperfusion. On the one hand, it was confirmed that the brain associates with thestomach and the intestine, and suggested that the brain must be treated as well as thestomach and the intestine in acute cerebral infarction clinical treatment. On the other hand,it provided an experimental basis and effective prescription for the clinical application ofactivating blood circulation and removing blood stasis in treating acute cerebral infarction.Methods:Copied80Middle Cerebral Artery Occlusion (MCAO) model rats. They were dividedrandomly into Tongqiao Huoxue Decoction conventional dose, low dose group, cerebralinfarction groups, and cimetidine group, who were given Tongqiao Huoxue Decoction,normal saline, cimetidine. In another20rats prepared sham operation group,20normalrats as normal control group were given normal saline. According to the rats were killed atdifferent times, the cerebral infarction group, sham operation group, the normal controlgroup would be divided into4d and7d two subgroups; Tongqiao Huoxue Decoction groupand the cimetidine group were sacrificed after the modeling4d and7d. There were12groups,10in each group. In the fasting state, after inferior vena venous blood samples were collected, the rats were decapitated, and the fresh gastrointestinal mucosas werecollected immediately. Venous blood samples were used to detect the gastrointestinalhormones content by radioimmunoassay; and the gastrointestinal mucosas were made tothe light and the electron microscopy section to observe their morphological changes.Results:1The contents of blood GAS, MTL, VIP in rats which get cerebral infarction wereincreased than the corresponding normal and sham operation group rats, there aredifferences (P <0.05); The contents of blood GAS, MTL, VIP in rats which in groupsTongqiao huoxue decoction convention, low dose and cimetidine were increased than thecorresponding normal and sham operation group rats, there are differences (P <0.05);Tongqiao huoxue Decoction conventional dose group could reduce the blood GAS, MTL,VIP contents more than low dose group and cimetidine group(P <0.05); Tongqiao huoxueDecoction conventional dose group and low dose group could reduce the blood GAS, MTL,VIP contents more than cerebral infarction group (P <0.05); The blood GAS, MTL, VIPcontent of cimetidine group showed no difference compared with Tongqiao huoxue lowdose group(P>0.05); The blood GAS, MTL, VIP content of normal group showed nodifference compared with sham operation group(P>0.05).2The contents of blood GAS, MTL, VIP in rats which get Tongqiaohuoxue Decoctionconventional dose group, low dose group and cimetidine7days group were decreased thanthe4d group (P <0.05); The contents of blood GAS, MTL, VIP in cerebral infarctiongroup, normal group and sham operation group rats changed no difference (P>0.05).3Cerebral infarction4d group could be seen visible gastric mucosal damaged in thenaked eye; under the light microscope, there would be easily found the gastrointestinalmucosa damaged, part of which could be found punctate hemorrhage, inflammatory cellswere infiltrated visibly, and sub mucosa had hyperemia or edema. Under the electronmicroscope, the mucus layer disappeared, the mucosa cells had wide range of ulceration,the cells arranged messily and exposed the basal layer, the form of gastric pits weredamaged. The injurious of the gastrointestinal mucosa4d subgroup are serious than the7dsubgroup.4The gastric mucosa of rats in Tongqiao Huoxue Decoction and cimetidine group didnot change significantly in the naked eye. Under the light microscope, gastric mucosalsurface epithelial structure was complete in Tongqiao Huoxue Decoction normal4d group,inflammatory cells’ infiltration was hardly to be seen, hyperemia and edema of sub mucosa could be seen mitigating than before. The injurious of the gastrointestinal mucosa4dsubgroup are serious than the7d subgroup. Inflammatory cells’ infiltration could be seen ingastric mucosal surface in low-dose Tongqiao Huoxue Decoction4d group, hyperemia andedema of sub mucosa could be seen mitigating than before. On4days, the damage ongastric mucosal surface of rats mitigated, inflammatory cells’ infiltration was hardly to beseen. The injurious of the gastrointestinal mucosa4d subgroup are serious than the7dsubgroup. Under the electron microscope, some areas of the rat gastric epithelial cellsruptured in Tongqiao Huoxue Decoction normal4d group, some areas of which exposedtheir basal layer, and the form of gastric pits were damaged. The injurious of thegastrointestinal mucosa4d subgroup are serious than the7d subgroup.Large areas of the ratgastric epithelial cells rupture in Tongqiao Huoxue Decoction low-dose4d group, whichexposed their basal layer, the form of gastric pits were damaged. The injurious of thegastrointestinal mucosa4d subgroup are serious than the7d subgroup.On4days, someareas of the gastric epithelial cells ruptured on rats which in cimetidine group, it could beseen that epithelial cells were flat, ruptured, and some had serious distortion, the form ofgastric pits were damaged.Conclusions:1There are pathological changes in the acute stage of cerebral infarction in rats’gastric mucosal morphology.2Early use of the Tongqiao Huoxue Decoction on gastrointestinal dysfunction in theacute stage of cerebral infarction has a protective effect.3The change of gastrointestinal hormone may be one of the main pathogenesis ofacute cerebral infarction with gastrointestinal dysfunction.
