Objective: Preliminary studied the plasma characteristic metabolites of patients withhyperactivity of Liver-Yang Sydrome in hypertension,to further study on substance ofsyndrome of hyperactivity of Liver-Yang sydrome inhypertension.Simultaneously,tosearch the role of Pingan Prescription and it’s composition as the target.Methods: Gas Chromatography-Mass Spectrometer (GC-MS) technology wasadopted in this experiment study to detect the plasma of patients with Hypertension ofsyndrome of ascendant hyperactivity of liver yang and syndrome of deficiency of liveryin-yang, and normal plasma.Simultaneously detected the plasma of spontaneouslyhypertensive rats(SHR) before and after the intervention of Pingan Prescription to findpatterns of metabolites. Respectively pretreatment the chromatogram and got thethree-dimensional date tables. By using the Pattern Recognition Technique of PrincipalComponent Analysis(PCA) and Partial least-square discriminant analysis(PLS-DA) tofind meaningful characteristic metabolites through PCA score plot. Then analysedchemical structures and metabolic pathways of those metaboliates to account for thebiologically significant of syndrome of ascendant hyperactivity of liver yang. Unscrambledthe characteristic metabolism and cleared out the essence of the syndrome of ascendanthyperactivity of liver yang. Results:9Metabolic biomarkers have been found through GC-MS technology inHypertension of syndrome of ascendant hyperactivity of liver yang, and their relatedmetabolic pathways have also been analysed. PCA showed that there were bothintersection and separate between Hypertension of syndrome of ascendant hyperactivity ofliver yang and deficiency of liver yin-yang, and the metabolic spectra of syndrome ofascendant hyperactivity of liver yang were more approach to the healthy adults. GC-MStechnology displayed that compared with the model group Ethanol-extraction group ofPinggan prescription had significant reduction of Stearic acid, Hexadecanoic acid, andArachidonic acid,but the Linoleic acid was increased obviously. The rest of the markershave no statistical significance.Conclusions: Metabonomics can be well applied in the exploration of the essence ofHypertension of syndrome of ascendant hyperactivity of liver yang, and evalution ofPingan Prescription. Metabonomics has validated the scientificity of the pathogenesisdevelopmental pattern of Hypertension of syndrome of ascendant hyperactivity of liveryang and deficiency of liver yin-yang, but also demonstrated that Pingan Prescription canimprove the metabolic spectra of SHR plasma effectively.
