BackgroudAdenomyosis is refers to the growth of the functional endometrial glands and stroma into the myometrium, and it is a benign disease while it has the biological behaviour similar to the malignant tumor. It mainly happened to the women at reprod-uctive age, and90%of the patients are multipara. About a third of the patients have atypical symptoms, and the most common symptoms of the other patients are the profuse menstrualtion (50%), period pain (30%), also including sexual pain, but also increase the possibility of abortion of the early pregnancy, and all of these symptoms are related to the depth and scope of the focus.Rho family has extensive biological effect. It can adjust the cytoskeleton restructuring to regulate the shape of the cells and influences the growth, proliferation and transfer of cells,and also can adjust the cell growth period which is related to the growth of cell differentiation. RhoE is a new member of the superfamily of the Ras guanine nucleotide binding protein, also known as Rnd3.The difference of the RhoE compared to the other members of the family of the Rho protein is that it only exists in the form of combination with GTP in the cells.At present, we realize that RhoE/Rnd3has the possibility to become an important tumor-suppressing gene, and analyze the relationship between the express of RhoE/Rnd3in the endometrial glandular cells and the clinical symptoms to discuss the function in the adenomyosis.Objective To investigate the expression of RhoE/Rnd3on the protein level in the eutopic endometrium and the smooth muscle tissues obtained from the patients without adenomyosis (control group) and the eutopic and ectopic endometrial tissues and the smooth muscle tissues surgically obtained from the patients with adenomyosis (experiment group) and its potential clinical effect.MethodsThe eutopic endometrium and the smooth muscle tissues obtained from the32patients in control group who have suffer from the hysterectomia because of uterine fibroids, ovarian cyst, uterine prolapse in provincial hospital affiliated shandong university from the January2010to march, and all of the tissues which are divided into proliferotive phase(14cases) and secretory phase(18cases) have been confirmed by pathological examination for the normal tissues. The eutopic and the ectopic endometrium and the smooth muscle tissues obtained from38patients postoperative identified by pathological examination with adenomyosis for the same period including20cases of proliferation.18cases of secretory. All of the patients were selected in three months without any hormone therapy and nervous system and endocrine system disease. The difference between the two groupes was no statistically significant (P>0.05). Immunohistochemical SP method was employed to detect the expression of RhoE/Rnd3on the protein level. The relationship of the express of RhoE/Rnd3between the endometrial glandular cells and the clinical symptoms was analysised,and then its function in the adenomyosis was discussed.Results1、In the endometrial glandular cells:RhoE/Rnd3protein located in the cytoplasm and membrance which appears light to brownish yellow, and the express strength in the membrane is greater than cytoplasm. The expression of RhoE/Rnd3in the control group were significantly higher than those of the eutopic(A groupe)(P<0.05) and the ectopic (B groupe)(P<0.05) endometrium of the experiment group. Statistical differences of the express of the RhoE/Rnd3were found in ecretory phase and estrin phase bettwen the control group and the experimental group A (P<0.05) and B (P<0.05).There are no statistical differences in the the eutopic and the ectopic endometrium glandular cells of the RhoE/Rnd3in the adenomyosis.2、In stromal and smooth muscle cells:there was no significant variance in the stromal cells of the three groups and the smooth muscle cells of the control group and the experiment group;3、The expression of RhoE/Rnd3in the endometrial glandular cells of the experiment group was related to course of disease of adenomyosis. The level of expression in the tissues obtained from the patients whose course had been exceeded two years were obviously lower than those had been not enough two years; but expression level of the RhoE/Rnd3protein has nothing to do with the dysmenorrheal.Conclusions1、The expression of RhoE/Rnd3in the control group were significantly higher than those of the eutopic (P<0.05) and the ectopic (P<0.05) endometrium of the experiment group,2、There was no significant variance in the stromal cells of the three groups and the smooth muscle cells of the control group and the experiment group.3、The expression of RhoE/Rnd3in the endometrial glandular cells was related to course of disease of adenomyosis. The level of expression in the tissues obtained from the patients whose course had been exceeded two years were obviously lower than those had been not enough two years.
