ObjectiveTo detect the levels of HSP60in gastric cancer (GC) tissue and cell lines, and analyze the relationships with clinicopathologic features and the roles in MVD. To investigate the potential role of plasma HSP60in the diagnosis and prognosis of GC.MethodsThe protein expression of HSP60were detected in85gastric cancer tissues and35normal adjacent tissues by immunohistochemistry respectively. Microvessel density(MVD) was evaluated by testing the expression of CD34. Twenty surgical specimens and corresponding adjacent normal gastric mucous tissues were collected and the expression of HSP60were confirmed by Real time PCR to illustrate the difference of HSP60between gastric cancer and corresponding adjacent normal gastric mucous tissues.The levels of HSP60in human gastric cancer cell lines AGS, BGC-823, SGC-7901, MKN-45, MKN-28and HGC-27were detected by Real-time PCR.HSP60was detected by ELISA in53cases with gastric cancer and46cases of health controls. At the same time, the diagnostic value of HSP60were analyzed.ResultsThe protein expression of HSP60was higher in gastric cancer tissues than in normal adjacent tissues (P<0.01), and the increased HSP60expression was Significant correlation with the size, differentiation and invasion of tumor (P<0.05), while the microvessel density in gastric cancer tissues was strong related to the size and invasion of tumor (P<0.05). In addition, significant association were seen between microvessel density and HSP60expression in gastric cancer tissues ((r=0.404, P<0.05))HSP60expression was examined in a panel of GC cell lines (AGS, BGC-823, SGC-7901, MKN-28, MKN-45and HGC-27) by quantitative RT-PCR. Interestingly, HSP60expression was strong in all these cell lines, which was different from that observed in normal adjacent tissues.The level of HSP60was significantly higher in gastric cancer group than that in health control group (P<0.001, respectively).. ROC curve analysis showed that AUC for HSP60was0.923. The results indicated that Plasma HSP60level could be a highly specific and sensitive index to diagnose gastric cancer.ConclusionThe higher expression of HSP60is significantly involved in angiogenesis and progression of gastric cancer, suggesting a role for HSP60as a prognostic biomarker and potential therapeutic target for gastric cancer.
