Objective To detect the expression of neuritin and HSP60 in traumatic liver injury tissues in a time-dependent manner in rats,and discuss the effect of neuritin and HSP60 on tissue repair of hepatic injury.Methods One hundred and twenty six SD rats were randomly divided into liver injury group(group A),fractures group(group B)and normal group(group C).In group A,the rat model of blunt liver injury was made by using modified BIM-IV biological impact machine.Group B make the right femoral fractures model in rats,and Group C was treated with blank.Each group divided into seven time points(6h,12 h,1d,3d,5d,7d and 14d),and the serum concentrations of ALT,AST,neuritin and HSP60 were respectively measured.Pathological changes of liver were observed by HE staining and immunohistochemical methods were used to detected the expression pattern of neuritin and HSP60 in liver tissue in rats.Result 1.42 rats of group A successfully made the model of blunt liver injury by using modified BIM-IV biological impact machine.2.The serum concentrations of ALT and AST in group A were higher than these in group B and group C(P<0.05)at the time points 6h,12 h and 1d.The serum concentrations of AST in group B increased at 6h,12 h and 1d,and it was statistically significant compared with group A and group C(P <0.05).3.HE staining of liver tissue in rats in group A showed hepatic lobules were destroyed.Hepatic sinus can be seen in a large number of red blood cells,liver cells edema,local necrosis,occasional mitotic figures,around hepatic sinusoid inflammatory cell infiltration.Then with the repairation of liver damage,red blood cells were significantly reduced in injured liver tissue and lymphocytes and neutrophil infiltration were observed.Meanwhile the number of endothelial cells increased.In the B group,the hepatic lobule structure of the rats in the fracture group was slightly disordered and the local cytoplasm was loose.4.The immunohistochemical staining results indicated the expression of neuritin and HSP60,at different time points from 12 h to 14 d,in group A is significantly higher than group B and group C(P<0.05).The expression of HSP60 in group B is higher than group C at the time points 12 h and 1d(P<0.05).And the expression of neuritin and HSP60 increased at first and then decreased.5.The serum Neuritin concentration of group A increased at 12 h,reached the highest value at 3d,then decreased gradually.In the fracture group(group B),the concentration of serum Neuritin increased gradually at 1 days,then gradually increased.Compared with group A,group B and group C at each time point,there was significant difference in serum Neuritin concentration between different time points(except 6 hours)(P<0.05).Liver injury group(group A)rat serum HSP60 was increased at 6h,12 h point reached a maximum value,then decreased gradually,the group B rat serum HSP60 concentration gradually increased slightly,then gradually decreased and down to the normal concentration at 3d.The serum HSP60 concentration of A group in 6h,12 h,1d were significantly higher than those in B group and C group(P<0.05).Conclusions Both neuritin and HSP60 were expressed in the injured liver tissues.With the change of time,the two show a consistent trend of change.The interaction between neuritin and HSP60 may play an important role in hepatic self-repairing after traumatic liver injury.
