Objective: This paper mainly study the expression and correlation of Twist and E-cadherinrespectively in osteosarcoma and osteochondroma, and evaluate the clinical significance ofthem in the occurrence, progression of osteosarcoma and osteochondroma.Methods: Expression of Twist and E-cadherin in50cases of osteosarcoma and12cases ofosteochondroma were detected by using immunohistochemistry method. Then, theexpression difference and clinical significance of Twist and E-cadherin were analyzed bySPSS16.0.Results:1. Twist mainly expressed in the cytoplasm of bone tumor cells. There were41positive cases in50osteosarcoma, the positive immunostaining rate was82.00%. Only2cases were positive in12osteochondroma, the positive immunostaining rate was16.67%.Which have significant difference(P<0.01). Expression of Twist have no obviousdifference in different pathological classification of the osteosarcoma(P>0.05), while havesignificant difference in different pathological grading of the osteosarcoma(P<0.05).2. E-cadherin mainly expressed in the cytoplasm of bone tumor cells, but a little expressionin the cytolemma. Have17cases were positive in50osteosarcoma, the positiveimmunostaining rate was34.00%, and9cases in12osteochondroma, the positiveimmunostaining rate was75.00%, which have significant difference(P<0.05). There is a nosignificant difference in different pathological classification and pathological grading ofthe osteosarcoma(P>0.05).3. The positive immunostaining rate of Twist in the transfer group of osteosarcoma was93.10%(27/29), and the no transfer group was46.15%(6/13). While, the positiveimmunostaining rate of E-cadherin in the transfer group of osteosarcoma was 24.14%(7/29), and the no transfer group was61.54%(8/13). Above two groups, which havesignificant difference(P<0.05). The expression of Twist was oppositely related withE-cadherin in osteosarcoma. There is a significant difference between them(rs=-0.457,P=0.001<0.01).Conclusion:1. The higer intensity of Twist protein is closely related to the formation anddevelopment of osteosarcoma. It can as a index to judge the garding of osteosarcoma.2. Osteosarcoma cells metastasis easily form the primary focal might be a defect inintercellular adhesion mediated by the absence of E-cadherin expression.3. Twist and E-cadherin can be used as auxiliary index of clinical prognostic ofosteosarcoma, so as to guide clinical treatment.4. Twist may promote invasion and metastasis of osteosarcoma by downexpression ofE-cadherin.
