Menopausal women with the decline of ovarian function,their ovarianfunction disorders, attenuates, reduces the secretion of estrogen, andovulation will gradually stop. Many women in the body cannot adapt to thechange of perimenopausal syndrome and appear a series of climactericsyndromes, which serious impacts on the woman’s physical and mentalhealth..Modern medicine mainly chooses symptomatic and hormonereplacement methods to control the climacteric syndrome, which ispositive.However it is not widely used with a limit for its contraindicationsand bid side effect. Acupuncture as an operation physical therapy is simple,safe and nontoxic side effects, efficacy, low cost in the prevention and itstreatment on perimenopausal syndrome has played a significant effect. TheGuanyuan as important points relating to the urinary tract, reproductivesystem disorders, which is selected as the main role in the acupuncture pointsacupuncture clinical research, and achieved remarkable results. However,acupuncture prevention of perimenopausal syndrome as a safe application on the limitations of physical methods has greater advantages anddevelopment potential, and is worthy of promotion and in-depth study. Thispaper, based on the roles of the acupuncture on human ovarian endocrineregulation,then choosing electricity and ultrasonic two physical methods,combining with the theory of traditional Chinese medicine (TCM) roled inthe guanyuan point. This paper, based on the acupuncture on human ovarianendocrine regulatory roles,used electricity and ultrasound two physicalfactors effecting in perimenopausal rats Guanyuan, to explore regulation ofwhich on the menopausal transition rats ovarian endocrine, and comparativestudy the influence of these two kinds of physical therapies on theperimenopausal rats ovarian endocrine and follicular development, analyzedelectrical stimulation and ultrasonic stimulation on the guanyuan ofmenopausal transition rat to improve the function of varian secretingestrogen and promoted feasibility of ovarian follicular development andfurther discussed the possible mechanism from which to provide basic datato the clinical application and research, which is of significance to developthe menopausal transition treatment in new ways.Objectives(1) Respectively, used electrical stimulation, ultrasound stimulation onthe Guanyuan of perimenopausal rats by observing the structural changes ofthe ovarian tissue in rats and serum estrogen levels to explore their effectionon secretion of estrogen function of ovarian and follicles development. (2) Via the electrical stimulation and ultrasonic stimulation on theguanyuan of the menopausal transition rat, detected expression of estrogenreceptorβin the ovary and cytochrome P450aromatase protein inimmunohistochemical way to preliminary discuss influence of electricalstimulation and ultrasonic stimulation on the guanyuan of acupuncturepoint.Methods(1) Screening out perimenopausal female SD rats60by Vaginal smearmethod,then randomly dividing into the electrical stimulation group(including electrical stimulation in30min, electrical stimulation in20mingroup, electrical stimulation in10min group f), ultrasound stimulation group(including ultrasound stimulation in10min group, ultrasound stimulation in5min) and model control group (n=10). Electrical stimulation groups:choose frequency2.5Hz, strength2V, continuous wave output, stimulate30min/d respectively,20min/d and10min/d; The ultrasound group: outputpower0.1W, working frequency9MHz, transmit frequenc1000Hz y,stimulate30s everytime, interval of5s, stimulate10min/d respectively and5min/d. The continuous stimulation after10days, having eyeball took blood,using the electrochemical luminescence immune analysis detected ratsserum estrogen level.(2) After electrical stimulation and ultrasonic stimulation continuousfunction on rat guanyuan10days, dislocated death rats after having taken blood,and had both sides for ovarian tissue slice.HE dyed histologic methodobservation ovarian histological structure change, count ovarian folliclesoriginal number, primary follicular number, subprime follicular number andatresia follicular number; Via immunohistochemical method detectionedovarian estrogen receptor beta and cytochrome P450aromatase proteinexpression.Results(1) After electrical stimulation and ultrasonic stimulation continuousfunction on rat guanyuan10days,the peripheral blood E level of the groupelectricity stimulated in30min is8.83.18±28.12pg/mL,the peripheralblood E level of the group electricity stimulated in20min is74.99.42±34.09pg/mL, the peripheral blood E level of the group electricity stimulated in10mins is54.00.±14.52pg/mL, the peripheral blood E level of the groupultrasound stimulated in10min is68.52±37.13pg/mL, the peripheral bloodE level of the group ultrasound stimulated in5mins is65.00±12.26pg/mL,the peripheral blood E level of the modern matched group is42.75±10.02pg/mL.From this we could conclude that The peripheral blood E level of themodern matched group is lower than any other groups while the groupelectricity stimulated in10mins is oppsite,which is of Statisticaldifferences(P<0.05); serum E levels of Electric experimental group andultrasonic experimental groups showed no significant difference (P>0.05);When electrical stimulation and ultrasound stimulation were compared between each other of the same experimental group, the serum E levels wereno significant differences (P>0.05).(2) After electrical stimulation and ultrasonic stimulation continuousfunction on rat guanyuan10days, Electrical stimulation in30min groupovarian follicles original for9.9+6.5, the primary follicular number is9.1+3.0, subprime follicular number is7.8+3.8, closure for its4.5+2.3;Electrical stimulation in20min group ovarian follicles original for8.7+4.2, the primary follicular number is8.6+2.6, subprime follicular number is7.0+4.0, closure for its4.8+3.1;electrical stimulation10min group ovarianfollicles original for8.1+3.8, the primary follicular number is6.1+2.4,subprime follicular number is5.4+2.2, closure for its6.0+2.0;Ultrasoundstimulate10in min group ovarian follicles original for7.9+4.5, the primaryfollicular number is7.1+2.0, subprime follicular number is6.8+1.8, closurefor its4.3+2.2; Ultrasound stimulate5min group ovarian follicles originalfor6.7+2.2, the primary follicular number is7.6+2.6, subprime follicularnumber is7.1+3.0, closure for its5.0+1.4;Model control group for ovarianfollicles original6.1+2.8, the primary follicular number is5.9+1.4,subprime follicular number is4.4+2.4, closure for its6.9+2.5. Among them,the electrical stimulation30min group, the electrical stimulation20mingroup and the ultrasonic stimulate10min group, the ultrasonic stimulate5min rats, ovarian primary follicular number, subprime follicle a modelcontrol group number significantly increased, atresia were far fewer follicles, there are significant differences (P <0.05), electrical stimulation10mingroup ovarian follicles, primary follicle original number number, subprimefollicular number and atresia follicle compared with the number is nostatistical difference (P>0.05). Comparing the Electrical stimulation withthe ultrasound stimulation group, the number of follicles at all levels, nosignificant difference (P>0.05). Electric experimental group, and ultrasonicexperimental group, the same experimental group between the two groupswere compared, still no significant difference in the number of follicles at alllevels (P>0.05). Electrical stimulation30min group, the electricalstimulation20min group and the ultrasonic stimulate10min group and theultrasonic stimulation5min rats, the expression of ovarian ER beta andcytochrome P450aromatase protein was significantly higher than the modelcontrol group (P <0.05), compared with the youth has no significantdifference (P>0.05). ovarian ER beta and cytochrome P450aromataseprotein expression of Electrical stimulation10min group was lower thanmodel control group a youth group, there exists significant difference (P <0.05).Conclusions(1) Electricity stimulated in guanyuan in30min/d and20min/d andultrasound stimulated in guanyuan in10min/d and5min/d both lasting10days,in which situation the electrical stimulation and ultrasonicstimulation could increase the level of the blood serum E and promote development of ovarian follicles growth.electrical stimulation in theguanyuan to increase the menopausal transition ovarian excretive estrogenfunction and promote the development of ovarian follicles is similar to to theultrasonical stimulation.(2) To improve the expression of the menopausal transition ovarianestrogen receptorβand cytochrome P450aromatase protein may be one ofthe mechanism that electrical stimulationand ultrasonical stimulationguanyuan to promote ovarian growth and follicular development.
