Objects:To detect the expression of Beclin1and PTEN in gastric caicinoma, andinvestigate its influence in carcinogenesis of gastric mucosa and prognosis.Methods:The tissue micioarrays containing199cases of gastric tumor specimens werefollowed up by telephone again in this study to get the survival data,while collecting15pairs of fresh gastric carcinoma and matched adjacent normal tissues for experimentalstudy.Using immunohistochemistry (IHC),Western blot to detect differentially expressedbetween the primary tumor and normal tissue,and investigate its influence in prognosis.Results: Immunohistoch-emistary show that overall expression rates of Beclin1and PTEN in cancer were47.2%(94/199)and55.8%(111/199),respectively. The lower expression of Beclin1and PTEN were associated with many clinicopathologicalfactors and prognosis. There was a significantly positive correlation between Beclin1and PTEN. Detected by Western blotting, the expression of Beclin1and PTENin carcinoma were significantly lower than that in the adjacent normal tissues.Conclusion: The lower expression of Beclin1and PTEN were associated with manyclinicopathological factors and prognosis,its influence in carcinogenesis of gastric mucosaand prognosis.The abnormal expression of Beclin1and PTEN may be related tocarcinogenesis and the development of gastric cancer.
