Background Bladder cancer is one of the most common clinical tumor in our department of Urology,and more than 90% were transitional cell carcinoma,PTEN(Phosphatase and tension homology deleted from chromosome 10)and autophagy gene Beclin1 transitional cell carcinoma in the bladder plays an important role in the development of PTEN,and Beclin1 may be an important marker of bladder transitional cell carcinoma and potential therapeutic target.Objective To detect the expression of PTEN and Beclin1 in bladder carcinoma tissues and normal bladder tissues and to analyze the role of the two in the development of bladder cancer.Methods Collect samples from 52 cases of bladder cancer patients(confirmed by pathological examination were transitional cell carcinoma),another 10 cases of normal bladder tissue as control,the expression in the tissue specimens by immunohistochemical detection of PTEN and Beclin1,bladder transitional cell carcinoma of the clinical and pathological factors combined to research the relationship between the two in bladder the expression and bladder cell carcinoma between transitional cell carcinoma and other biological characteristics.Results 1.The positive expression rate of PTEN in bladder transitional cell carcinoma and normal bladder tissue was 53.84% and two respectively,and the difference was statistically significant(P<0.05).2.The positive expression rates of Beclin1 in bladder transitional cell carcinoma tissue and adjacent normal bladder tissue were 59.62% and two respectively,and the difference was statistically significant(P<0.05).3.Expression and histopathological grades,PTEN staging was significantly related to TNM(P<0.05),but not with age and gender was significantly correlated(P>0.05);expression and pathological grades,Beclin1 tumor TNM stage(P<0.05),and was significantly associated with gender,age had no significant correlation(P>0.05).4.In transitional cell carcinoma of the bladder,the positive expression rates of PTEN and Beclin1 were positively correlated(r=0.378,P<0.05).Conclusion 1.Compared with normal bladder tissue,the expression of PTEN in bladder transitional cell carcinoma was significantly lower than that in normal bladder transitional cell carcinoma,and it was significantly correlated with the histological grade and TNM stage of the tumor(P<0.05).2.Compared with normal bladder tissue,the expression of Beclin1 in bladder transitional cell carcinoma was significantly decreased,and it was significantly correlated with the pathological grade and TNM stage of the tumor tissue(P<0.05).3.There was positive correlation between the expression of PTEN and Beclin1 in bladder transitional cell carcinoma.PTEN and Beclin1 decrease to promote the occurrence and development of tumor,and the joint detection has a certain significance.
