Objective: To investigate the relations between the human leucocytesantigen-DQA1(HLA-DQA1) alleles polymorphism and the Uygur、Han nationalityhepatitis B patients in Xinjiang and the genetic susceptibility healthy controls.Which isprovide some important clues to seek the susceptible genes and disease~resistant genes ofHBV infection for Uygur and Han nationality hepatitis B patients. Methods:(1) Collect0.6ml peripheral blood of every subject, extract peripheral blood genome with extractreagent of Sango Biotech Co. Ltd, then check the DNA quality use0.7%agarose gelelectrophoresis and nucleic acid protein measuring apparatus;(2) PCR-SSP was used toanalyze the frequency distribution of HLA-DQA1*0102*0104*0201*0301*0302*0501alleles among of Han nationality and Uygur patients with HBV and Han nationality andUygur healthy controls.(3) Take the PCR product6uL fully blended with bromorphenolblue2uL, using1.5%agarose gel electrophoresis (contain EB). Then observe the result bythe ultraviolet ray transmission gel imager and capture the images.(4) Statistics analyseswere analyzed by SPSS17.0software, HLA-DQA1allele’s frequency (AF) using directedtechnology method and using2×2four cell tableχ~2test or according Fisher definiteprobability method, P<0.05indicate have obvious difference[1-2]. Results:(1)HLA-DQA1*0501allele gene frequency expression is lower than the Uygur nationalitynormal healthy controls groups χ~2=6.91, P<0.05,OR(95%CI),respectively, in:0.38,(0.18~0.79) difference have statistic significance.(2) Compared the Han nationality hepatitis Bpatients ALT abnormal group with healthy controls group in allele’s frequency analysisfound that HLA-DQA1*0102*0201*0301sites genes respectively P<0.05,OR(95%CI)were3.75(1.588~8.853)、0.28(0.119~0.662)、4.67(1.811~12.028)difference have statisticsignificance.(3) Compared the Han nationality hepatitis B patients ALT normal groupwith healthy controls group in allele’s frequency analysis found that HLA-DQA1*0201sites genes P<0.05,OR(95%CI) were0.150(0.061~0.357),difference have statistic significance.(4) Compared the Uygur hepatitis B patients ALT abnormal group withhealthy contrast group in allele’s frequency analysis found thatHLA-DQA1*0301*0501genes respectively P<0.05OR(95%CI)were7.52(3.379~16.750)0.01(0.001~0.081),difference have statistic significance.The result same to analysisfounder between the Uygur hepatitis B patients ALT normal group with Uygur healthycontrast group.(5) Compared the Han nationality hepatitis B patients HBV high copyquantity group with healthy controls group in allele’s frequency analysis found thatHLA-DQA1*0102*0201*0301*0501genes respectively P<0.05,OR(95%CI) were4.67(1.947~11.185)0.16(0.064~0.417)3.79(1.473~9.763)8.98(3.579~22.551),differencehave statistic significance.(6) Compared the Han nationality hepatitis B patients HBV lowcopy quantity group with healthy controls group in allele’s frequency analysis found thatHLA-DQA1*0201genes P<0.05,OR(95%CI) were0.23(0.104~0.529), difference havestatistic significance.(7) Compared the Han nationality hepatitis B patients ALT normalgroup with Han nationality hepatitis B patients ALT abnormal group in allele’s frequencyanalysis found that HLA-DQA1*0102*sites genes P<0.05,OR(95%CI) were0.27(0.103~0.698)difference have statistic significance.(8) Compared the Uygurhepatitis B patients HBV high copy quantity group with healthy contrast group in allele’sfrequency analysis found that HLA-DQA1*0301*0501genes respectivelyP<0.05,OR(95%CI)were8.21(3.621~18.604),0.02(0.005~0.098)difference have statisticsignificance;This same to analysis founder between the Uygur hepatitis B patients HBVDNA low copy quantity group with Han nationality.Conclusion:(1) HLA-DQA1*0501is the protected gene of Uygur hepatitis B patients.(2) The Han nationality hepatitis bpatients group HLA-DQA1*0102*0301and the abnormality of ALT level have positivecorrelation;HLA-DQA1*0201and the abnormality of ALT level have negativecorrelation;the Han nationality hepatitis b patients group HLA-DQA1*0102*0301*0302*0501has positive correlation with HBV DNA copy level;HLA-DQA1*0201has negative correlation with HBV DNA copy level.(3) The Uygur hepatitis b patientsgroup HLA-DQA1*0301and the abnormality of ALT level and HBV DNA copy levelhave positive correlation; HLA-DQA1*0501and the abnormality of ALT level and HBVDNA copy level have negative correlation.
