Objective: To investigate the association between polymorphism of F+1, V4 locus in ADAM 33 gene and chronic obstructive pulmonary disease(COPD) and lung function in Xinjiang Uygur and Han population.Methods: Blood samples from 200 COPD patients and 211 healthy controls in smoking Uygur population and 199 COPD patients and 210 healthy controls in smoking Han population were collected.Samples of DNA was extracted, and F+1, V4 single nucleotide polymorphism(ADAM 33) was detected by ABI SNa Pshot SNP genotyping.Using SPSS 20.0 statistical software for statistical analysis of all data,statistical analysises adopt One-way ANOVA, Logistic regression analysis, t test, χ2 test, and calculate the values of odds ratio and 95% confidence intervals for odds ratio. Results: 1. Case groups and control groups general information were compared display: two groups of gender(male, female), age(years),smoking index(number of cigarettes smoked per day×years of smoking, cigarettes/year), BMI(body mass index, BMI, kg/m2), the differences were not statistically significant(P> 0.05); Lung function indices(FEV1 predicted value(%), FEV1/FVC) compared, the differences were statistically significant(P<0.05). 2. Case groups and control groups ADAM33 gene F+1,V4 locus genotypes distribution consistent with Hardy-Weinberg genetic equilibrium law(P> 0.05);3. On the ADAM33 gene F+1,V4 locus, genotype constitute than of cases group(F+1 locus C/C,C/T,T/T genotypes were : Uygur 45.0%,44.0%,11.0%;Han 52.3%,38.7%,9.1%; V4 locus G/G,G/C,C/C genotype were: Uygur 43.5%,45.5%,11.0%;Han 39.7%,44.2%,16.1%); genotype constitute than of control group(F+1 locus C/C,C/T,T/T genotypes were : Uygur 47.9%,44.5%,7.6%;Han 53.8%,38.1%,8.1%; V4 locus G/G,G/C,C/C genotype were:Uygur 48.8%,43.1%,8.1%;Han 42.4%,43.8%,13.8%), Comparison of ADAM33 gene F+1,V4 locus genotype frequencies distribution, the difference was not statistically significance(Uygur F+1:χ2=1.485,P=0.476;V4:χ2=1.695,P=0.428;Han F+1:χ2=0.163,P=0.922;V4:χ2=0.536,P=0.765). 4. On the ADAM33 gene F+1,V4 locus, allele frequencies constitute than of cases group(F+1 locus C, T allele were: Uygur 67.0%,28.4%;Han 71.6%、28.4%;V4 locus G, C allele were: Uygur 66.3%,33.8%;Han61.8%,38.2%); allele frequencies constitute than of control group(F+1 locus C, T allele were: Uygur70.1%,29.9%;Han 72.9%,27.1%;V4 locus G, C allele were: Uygur 70.4%、29.6%;Han 64.3%、35.7%),Comparison of ADAM33 gene F+1, V4 locus allele frequencies distribution, the difference were not statistically significance(Uygur F+1:χ2=0.941,P=0.332; V4:χ2=1.619,P=0.203;Han F+1:χ2=0.159,P=0.690;V4:χ2=0.538,P=0.463). 5. Association between cases group F+1,V4 locus genotype and clinical indicators of lung function display: there were no significant differences in F+1,V4 locus genotype and clinical indicators of lung function display, and in the FEV1% predicted and FEV1/FVC in Uygur population((F+1:F=0.595,P=0.552;F=0.676,P=0.510;V4:F=0.391,P=0.677;F=1.562,P=0.212)).There were significant differences in V4 GG genotype and clinical indicators of lung function display, and in the FEV1% predicted and FEV1/FVC in Han population(F=4.735,P=0.010;F=5.336,P=0.006). There were no significant differences in F+1 locus genotype and clinical indicators of lung function display, and in the FEV1% predicted and FEV1/FVC in Han population(F=1.852, P=0.160;F=0.830, P=0.438).Conclusion: 1. There is no significant difference in the polymorphism of F+1, V4 locus in ADAM 33 gene and the susceptibility to COPD in Xinjiang Uygur and Han population.2. Patients with V4 locus had much lower predicated value of FEV1% predicted and FEV1/FVC than other genotype in Han population.
