0bjective: To investigate the effect of artesunate on expression of the T cell-specifictranscription factors T-bet/GATA-3in oral lichen planus (OLP).Methods: Peripheral blood mononuclear cells (PBMC) were isolated by densitygradient centrifugation from OLP patients.The cells were treated with differentconcentration of artesunate(0、1、100μmol/L) and5%sodium bicarbonate injectionfor24hours. Expressions of T-bet and GATA-3mRNA and protein in the PBMCswere detected by reverse transcription-polymerase chain reaction (RT-PCR) andWestern Blot respectively.Results: The expressions of T-bet mRNA and protein in100μmol/L artesunate(0.54±0.26、0.93±0.40)were lower than in the control group(1.35±0.63、1.47±0.43)(P<0.01、P<0.01). The expressions of GATA-3mRNA and protein in1μmol/L(0.62±0.33、1.11±0.69)and100μmol/L(0.23±0.21、0.44±0.27)artesunatewere both lower in the control group(1.40±0.66、1.68±0.61) significently(P<0.01、P<0.01、 P<0.01、P<0.01).Conclusions: Artesunate could rectificate the imbalance of transcription factors T-betand GATA-3. Artesunate may privode a thread in the treatment of OLP. |
