Objective:The infiltration of the inflammatory cells in human nasal polyps and EOS in human nasal polyps were observed by HE staining, and the expression of IL-17, IL-4and IFN-y in nasal polyps were detected by SABC immunohistochemistry. To analyze the role of the ratio imbalance of Thl/Th2/Th17cells in pathogenesis of nasal polyps.Materials and Methods:Materials:30inpatients (17males and13females, age range is from17to60, average age is41.7) who were in Yanbian University Hospital from December,2009to December,2012with chronic sinusitis (the degrees of nasal polyps are above Ⅱ degree) were selected as the nasal polyps group and all the inpatients (no previous history of nasal polyps, allergic rhinitis, asthma and other allergy and systematic diseases) were treated with endoscopic surgery in Yanbian University Hospital. Nasal polyps specimens are verified by more than two associate chief physician of department of Pathology.20inferior turbinate mucosa from the inpatients (no previous history of allergic rhinitis and systematic diseases) treated with nasal septal reconstruction were selected as the control group (12males and8females, age range is from18to53, average age is31.6).Methods:50formalin fixed and paraffin embedded tissues were stained by HE and SABC immunohistochemistry. The infiltration of the inflammatory cells and EOS were observed by HE staining. And the expressions of IFN-γ, IL-4and IL-17are detected with the use of SABC immunohistochemistry. The data from the two kinds of tissues are expressed as mean±and standard deviation (x±S) with T-test. The expressions of IFN-γ、IL-4and IL-17in the two kinds of tissues are detected by SABC immunohistochemistry. The expressions of the three kinds of cytokines in two kinds of tissues are expressed as mean±and standard deviation (x±S) with T-test. Correlation between EOS and the three kinds of cytokines is analyzed by using Pearson line.Results:1. The infiltration of eosinophil in nasal polyps group was significantly higher than the normal inferior turbinate group (p<0.01).2. There is no significant difference between IFN-y in nasal polyps group and normal inferior turbinate group (p>0.05).3. The expression of IL-4in nasal polyps group was significantly higher than the normal inferior turbinate group (p<0,01).4. The expression of IL-17in nasal polyps group was significantly higher than the normal inferior turbinate group (p<0.01).5. EOS and IFN-gamma in nasal polyps lack of correlation, the correlation coefficient r=-0.206(p>0.05).6. There is a positive correlation between EOS and IL-4in nasal polyps, the correlation coefficient r=0.392(p<0.05).7. There is a positive correlation between EOS and IL-17in nasal polyps, the correlation coefficient r=0.421(p<0.05). Conclusion:The expression of IL-4and IL-17in nasal polyps were significantly enhanced, suggeesting that nasal polyps the Th2/Thl7cell dominant. Large number of EOS are detected in nasal polyps, IL-4and IL-17may be involved in chemotaxis and aggregation of the EOS, and play an important role in process of nasal polyps.
