Objective:Liver-depression-spleen-deficiency syndrome of hepatitis animal model was made by subcutaneously injected with CCl4and chronic restraint. Improved Prescription of Carefree Pill Decoction was used for the above animal model and the methods of liver-depression-spleen-deficiency syndrome of chronic hepatitis was preliminarily formed and the comprehensive efficacy evaluation index was establishedMethods:Thirty-two adult male rats, weighted250g to300g, were used in the study. The rats were divided into A group (blank group group, including eight rats), B group (model group, including twelve rats), and C group (Improved Prescription of Carefree Pill Decoction group, including twelve rats) after adaptive breeding for one week. All the twenty-four rats were subcutaneously injected with10%CCl4twice one week for four weeks, according to0.5ml/100g and the first time used twice dose.ALT and AST were tested for two rats, which were randomly selected from the model group to determine the degree of liver damage. After one week of modeling, chronic stress model was established by blinding up the rats of group B and group C into special tubes from eight a.m. to eleven a.m. for twenty-one days.Chronic restraint was not used in group A, but fasting was conducted at the same time of group B and group C for twenty-one days. Rats of group C was adiministered by intragastric administration of Improved Prescription of Carefree Pill Decoction for three weeks, using thirty times dose than patient dosage, thirty minutes before chronic restraint after one week of the experiment. Rats of group A was adiministered by equal distilled water. The emotional behaviours, daily consuming amounts,diarrhea, and autonomous activity of experimental animal was observed. D-xylose Excretory Rate was tested after the rats were gavaged with D-xylose every week. And the change of5-HT, NE,DA, ALT, AST, SS, MTL, VIP, TBILI, DBILI, TP, ALB and the liver pathologic change were observed. T test was used for the with-group comparisons of the above index. Results:Diarrhea and rattiness, and body weight was decreased appeared in the experimental animal and the dietary and drinking amounts also decreased. The index of liver, spleen, and adrenal all decreased of the Improved Prescription of Carefree Pill Decoction group. Compared to model group, the D-xylose contents of blank group group and Improved Prescription of Carefree Pill Decoction group both lower. And the5-HT and DA contents significantly increased (P<0.05;P<0.01) while the NE contents showed decreasing trend. The level of ALT and AST both significantly lower than model group. The ALB and TP increased (P<0.001; P<0.05) and the DBLI showed decreasing trend. There were no significant differences of TBILI among the groups. The level of MTL, VIP, and SS all significantly increased (P<0.05; P<0.001; P<0.05). The level of IL-2significantly decreased (P<0.05) while IL-6, and IL-8both showed decreasing trend. The hepatic tissue structure of the blank group group was normal while the hepatic lobules structure of the model group was not clear and patchy necrosis can be observed. The hepatic lobules structure of the Improved Prescription of Carefree Pill Decoction group was normal but obvious cloudy swelling ballooning degeneration or fatty degeneration was observed and sporadic dot necrosis appeared.Conclusion:The comprehensive efficacy evaluation index of liver-depression-spleen-deficiency syndrome of hepatitis animal model was established using the above experimental methods.
