Objective To investigate the possible role of adenosine and cytokines in pathogenesis of acute bacterial prostatitis and to explore new way to the treatment of acute bacterial prostatitis, by studying the concentration of adenosine and the expression of TNF-α and IL-10in rat prostate tissue. Methods After building the rat model of acute bacterial prostatitis, the prostate tissue were collected in4days and14days, the expression of TNF-α and IL-10in the prostate were detected by using immunohistochemical techniques. The data were derived by using computer image analysis system (Image-Pro Plus6.0) to measure the mean optical density (MOD) of the positive region. The concentration of adenosine was detected by using high performance liquid chromatography. The data were statistically analyzed with the SPSS11.0software.Results (1) The concentration of adenosine and the expression of TNF-α and IL-10in tissue of acute bacterial prostatitis were significantly higher than that in the control group.(2) Adenosine receptors were blocked by theophylline, the concentration of adenosine and the expression of TNF-α in the rat prostate tissue were significantly higher than that in the control group; but the expression of IL-10was significantly lower than that in non-intervention group. Conclusions (1) Adenosine concentration was increased, the expression of TNF-α and IL-10were enhanced in acute bacterial prostatitis. It shows that adenosine, pro-inflammatory cytokines and trans-inflammatory cytokines probably participate the pathogenesis of acute bacterial prostatitis, and immune factors may play an important role in acute bacterial prostatitis.(2) The imbalance of pro-inflammatory factors and anti-inflammatory factors may be the important pathogenesis of acute bacterial prostatitis.(3) Non-selectively blocking the adenosine receptors make adenosine concentration increase in prostate tissue, adenosine may play some kinds of role in pathogenesis of acute bacterial prostatitis.
